西黄丸对荷瘤大鼠免疫功能的影响

摘　　要：目的:研究西黄丸乙醇提取物对荷瘤大鼠模型免疫功能的影响。方法:用W256乳腺癌细胞建立荷瘤大鼠模型,随机分为7组:正常对照组、模型对照组、阳性药物组(香菇多糖230.0mg/kg)、西黄丸高剂量组(1.37g/kg)、西黄丸乙醇提取物高(65.8mg/kg)、中(26.3mg/kg)、低(13.2mg/kg)剂量组。造模后第二天起连续治疗14天,腹主动脉取血,采用流式细胞仪检测外周血中T淋巴细胞亚群含量;采用ELISA检测外周血中IL-2、IL-10、IFN-γ的表达水平。结果:西黄丸乙醇提取物高(65.8mg/kg)、中(26.3mg/kg)剂量组可提高荷瘤大鼠外周血中IL-2、IFN-γ的表达水平和CD3+、CD4+、CD8+的细胞比例,降低IL-10的表达水平,与等量的西黄丸比,其原制剂乙醇提取物优于西黄丸。结论:西黄丸乙醇提取物通过改善荷瘤机体免疫功能,调节肿瘤相关炎性因子的表达,发挥抗肿的瘤作用。Objective： To explore the T lymphocyte subsets and inflammatory effect of Xihuang Pill ethanol extract on tumor-bearing rats.Methods： W256 breast cancer models were established in rat divided into 7 groups（n = 10） ： normal group;model control group;lentinan group（23mg/kg）;Xihuang Pill high group（1.37g/kg）;Xihuang Pill ethanol extract（13.2mg/kg）;middle Xihuang Pill ethanol extract（26.3mg /kg）;high Xihuang Pill ethanol extract（68.5mg / kg）.Continuous treatment for 14 days,taken blood from abdominal aorta,CD3＋、 CD4＋、CD8＋in peripheral blood was detected by Flow Cytometer（FCM）,IL-2,IL-10,IFN-γ in peripheral blood was detected by ELISA.Results： The level of IL-2,IFN-γ,CD3＋,CD4＋,CD8＋in model control group was significant lower than the high（68.5mg / kg） and middle（26.3mg/kg） Xihuang Pill ethanol extract group（P ＆lt;0.05）.The level of IL-10 in model control group was significant higher than the high（68.5mg/kg） and middle（26.3mg/kg） Xihuang Pill ethanol extract group（P ＆lt;0.05）.The Xihuang Pill ethanol extract indicator was better than Xihuang Pill in equivalents dose.Conclusion： Xihuang Pill ethanol extract can improve the bioavailability of Xihuang Pill.The Xihuang Pill ethanol extract can improve the immune function and regulate the inflammatory effect.
