摘 要:目的:研究环维黄杨星D(CVB-D)对大鼠胸主动脉内皮完整血管环的舒张作用,并探讨其可能的作用机制。方法:采用80 mmol/L氯化钾(KCl)或1μmol/L去氧肾上腺素(PE)预收缩血管,观察CVB-D在给药浓度范围(1×10-5~6×10-4mol/L)对血管的舒张作用;将6×10-4mol/L CVB-D与血管预孵育,观察其对KCl(10~80 mmol/L)或PE(1×10-9~1×10-6mol/L)收缩血管作用的影响;通过观察不同抑制剂对CVB-D舒张大鼠离体血管作用的影响,考察NO途径、前列环素(PGI2)途径以及钾离子通道与内皮依赖的血管舒张作用之间的关系。结果:在给药浓度范围(1×10-5~6×10-4mol/L)内,CVB-D对80 mmol/L KCl或1μmol/L PE预收缩血管环的舒张作用呈剂量依赖性;6×10-4mol/L CVB-D与血管预孵育可抑制KCl(10~80 mmol/L)或PE(1×10-9~1×10-6mol/L)引起的血管收缩;0.3 mmol/L L-NAME或30μmol/L ODQ均可阻断CVB-D的血管舒张作用,吲哚美辛、4-氨基吡啶、格列苯脲以及四乙铵对CVB-D的舒血管作用没有影响。结论:CVB-D对大鼠离体胸主动脉的血管舒张作用呈剂量依赖性,其内皮依赖的舒张作用可能与NO-GC-cGMP途径相关。Objective: To observe the vasorelaxant effects of cyclovirobuxine D on isolated rat thoracic aorta rings with endothelium and to investigate the possible mechanism.Method: The vasodilation of cyclovirobuxine D at various concentrations(range from 1 × 10-5to 6 × 10-4mol / L) on 80 mmol / L potassium chloride(KCl) or 1 μmol / L phenylephrine(PE)-precontracted aorta rings were observed.KCl(10 ~ 80 mmol / L) or PE(1 × 10-9~ 1 × 10-6mol / L)-induced contraction was also recorded after the aorta rings were preincubated with CVB-D at the concentration of 6 × 10-4mol / L.When the aortic rings were in incubated with different inhibitors,the effect of CVB-D on aortic rings was examined and investigated.Results: To KCl or PE-precontracted aorta rings,CVB-D showed dose-dependent vasorelaxant effect at the given range of concentrations(1 × 10-5~ 6 × 10-4mol / L) on the rings preconstricted by KCl(80 mmol / L) or PE(1 μmol / L).CVB-D(6 × 10-4mol / L) preincubation could inhibit KCl(10 ~ 80 mmol / L) or PE(1 × 10-9~ 1 × 10-6mol / L)-induced contraction,additionally,NO synthase inhibitor(LNAME)(0.3 mmol / L) or selected NO sensitive soluble guanylate cyclase inhibitor(ODQ)(30 μmol / L) block the vasodilation effect of CVBD.While Indo,4-AP,Gly and TEA showed little effect on CVB-D.Conclusion: CVB-D has effect on relaxation of rat isolated thoracic arterial rings in a dose-dependent manner,the endothelium-dependent vasodilation may be related to the NO-GC-cGMP pathway.
