毛脚鵟气管源细胞分离及体外培养

摘　　要：研究了毛脚鵟气管源细胞的体外培养,分析了培养液、血清、胰岛素、肌苷等一系列因素对细胞生长增殖的影响。采用组织块贴壁法成功培养了毛脚鵟气管源细胞,用四种不同的基础培养液(DMEM(高糖)、DMEM/F12、M199、MEM alpha)对细胞进行培养,结果显示DMEM/F12对毛脚鵟气管源细胞的培养效果最好。毛脚鵟气管源细胞不具血清依赖性,10%血清的培养基中培养为宜。添加一定浓度的胰岛素、肌苷均能促进细胞的生长。Tissue culture of cells from trachea tissue of Rough-legged Buzzard and the effects of a series of factors such as medium,serum,insulin,inosine on the cell culture were investigated in the current study.Cells from trachea tissue of Rough-legged Buzzard were obtained successfully by standard tissue culture methods.Four kinds of mediums （DMEM（high glucose）,DMEM/F12,M199,MEM alpha） were used during the process of subculture.The result showed that DMEM/F12 was the optimal medium for the cell culture.The cells did not present a serum dependence,but the cell could grow well in the medium containing 10％ serum.Certain concentration of insulin and inosine could also promote the cell culture.
