摘 要:【目的】筛选杂交相思组织培养不同培养阶段的培养基配方,为实现杂交相思工厂化育苗提供技术参考。【方法】用杂交相思嫩茎为外植体,探究生长调节剂6-BA、NAA、IBA对其诱导出芽、继代增殖和诱导生根的效果。【结果】在不添加6-BA的情况下,添加0.2mg/LNAA时芽诱导率为60.0%,当添加0.2mg/L的6-BA后,诱导率为42.0%。当NAA为0~0.5mg/L时,随着6-BA的增加,增殖系数随之增加,6-BA为0.8mg/L时,增殖系数为4.2;当6-BA为1.0mg/L以上时,继代苗的增殖系数为1.0。以改良MS为基本培养基,在NAA1.0mg/L的条件下添加0.5、1.0mg/L的IBA生根诱导率分别为17.5%、24.0%;以改良1/2MS为基本培养基,在NAA1.0mg/L的条件下,添加0.5mg/L的IBA生根率为91.7%,当添加IBA为1.0mg/L时,生根率降到50.0%;生根苗移栽基质以黄心土与河沙或蛭石混合较好,移栽成活率在85.0%以上。【结论】改良MS+NAA0.2mg/L培养基是较适宜芽诱导的培养基;改良MS+6-BA0.8mg/L+NAA0.5mg/L培养基较利于继代苗的生长;改良1/2MS+NAA1.0mg/L+IBA0.5mg/L培养基较改良MS培养基有利于诱导生根。[Objective]The rapid propagation technique of Acacia auriculaeformisxAcacia mangium was studied to screen proper culture medium formulation for different stages and to achieve industrializing breeding. [Method]Using hybrid Acacia stems as explants for tissue culture, effects of growth regulators including 6-BA, NAA and IBA of different concentrations on induced sprouting, subculture propagation and root induction were explored. [Result]The results showed that in case of 0.2 mg/ L NAA, the initial induction rate was 60.0~/o when no 6-BA was added, and the induction rate was 42.0% when there was0.2 mg/L 6-BA. At 0- 0.5 mg/L NAA, proliferation coefficient increased with the increase of 6-BA concentration. At 0.8 mg/L 6-BA, the propagation coefficient was 4.2. When 6-BA was above 1.0 rag/L, transgenerational seedling proliferation coefficient was 1.0. Using the improved MS as the basic medium, under the condition of 1.0 mg/L NAA, rooting induction rate was 17.5% and 24.00/o for 0.5 and 1.0 mg/L IBA, respectively. Using the 1/2 modified MS as the basic medium, under the condition of 1.0 mg/L NAA, rooting rate was 91.7% and 50~/o for 0.5 mg/L IBA and 1.0 mg/L IBM respectively. When rooting seedlings were transplanted in substrate with yellow subsoil and river sand or vel^niculite mixture, transplanting survival rate was more than 85.0%. [Conclu- sion]Improved MS +NAA 0.2 mg/L medium was more appropriate for the initial induction medium. Improved MS+6-BA 0.8 rag/ L+NAA 0.5 mg/L was conducive to the growth of transgeneratioual seedlings. 1/2 modified MS+NAA 1.0 mg/L+IBA 0.5 mg/L was better for root induction.
