体外培养破骨细胞功能的定量评定

摘　　要：目的　建立适用于体外药理学研究的破骨细胞骨吸收功能的定量评定方法。方法　机械分离法获得破骨细胞 ,接种于象牙簿片底物上 ,分别于培养 1d、3d、5 d、7d取出象牙簿片各 4张 ,采用光镜和扫描电镜观察骨吸收陷窝数量的变化 ;对随机拍摄的陷窝照片进行计算机形态计量分析 ,测算陷窝的面积和深度。结果　骨吸收陷窝计数光镜法和扫描电镜法具有良好的相关性 ,r=0 .9998,P<0 .0 0 1。随着培养时间的延长 ,陷窝数量逐渐增多、面积扩大、深度加深。结论　骨吸收陷窝计数光镜法可以用于体外培养破骨细胞功能的定量评价 ,结合陷窝面积和深度分析 。Objective To establish a quantitative evaluation method of absorptive function of osteoclastwhich is fitforpharm acological studies.Method Get osteoclast by mechanical dispersion method,inoculate it on thin ivory slices,take outfour slices respective- ly after culturing for1d,3d,5 d,and7d,observe the changes of the lacuna am ountof bone absorption with lightmirror and scan- ning electric mirror,make a quantitative analysis of computer form to the random ly taken pictures of lacunae,measure the area and depth of the lacunae.Results L ight mirror method of bone absorptive lacunae amount had a good correlation with scanning electric m irror m ethod,r=0 .9998,P<0 .0 0 1.With increasing the culture tim e,there are more,larger and deeper lacunae.Con- clusions Counting light mirror method of bone absorptive lacunae could be used for quantitative evaluation of the osteoclast function cultured in vitro,combining with the analysis of lacunae area and depth,we could more completely evaluate the bone absorptive function of osteoclast cultured in vitro.
