摘 要:目的研究重度烧伤患者外周血T淋巴细胞亚群及血清TGF-β含量的变化,探讨患者血清TGF-β含量与免疫功能变化的临床意义。方法采用免疫荧光流式细胞技术检测了45例重度烧伤患者及45例正常成人外周血T淋巴细胞亚群CD3+、CD4+、CD8+细胞水平及NK细胞活性;采用ELISA方法检测了上述重度烧伤患者及正常成人血清TGF-β含量。结果重度烧伤患者CD3+、CD4+、CD8+细胞百分比均低于对照组(P<0.01);重度烧伤患者NK细胞活性(7.11±4.25)明显低于对照组(14.90±4.91)(P<0.01);而重度烧伤患者血清TGF-β含量(9.19±4.26)μg/L明显高于对照组的(4.49±1.91)μg/L(P<0.01)。结论重度烧伤患者T淋巴细胞亚群及NK细胞比例改变,提示重度烧伤患者存在免疫功能异常,而血清TGF-β含量与重度烧伤患者免疫功能密切相关,为进一步揭示重度烧伤患者损伤机制提供理论依据。OBJECTIVE To study on the changes of T lymphocyte subtype levels and the contents of serum transforming growth factor 13(TGF-β) in severe burn injurys patients so as to discuss the clinical significance of the change of serum TGF-β and the immune function. METHODS The flow cytometry (FCM) was used to detect the T lymphocyte subtypes including CD3- , CD4+ , and CD8+- and NK cells in 45 cases of patients with severe burn inju- ries; ELISA was used to detect the changes of TGF-β in the severe burn injurys patients and the healthy adults. RESULTS The study showed that the percentages of CD3+ , CD4+ , and CD8+ cells of the severe burn injurys patients were all lower than that of the control group (P〈0.01), the activity of the NK cells of the severe burn injurys pa- tients was (7.11±4.25), significantly lower than (14.90±4.91) of the control group (P〈0.01) ; the content of the serum TGF-13 of the severe burn injurys patients was (9.19t:4.26) /1g/L, significantly higher than(8.07±1. 91) bcg/L of the control group (P〈0.01). CONCLUSION The change of the ratio of T lymphocyte subtype and NK cells indicates that there is immune dysfunction of the severe burn injurys patients, and the serum TGF-β con- tent is closely related to the immune function, which provides the theoretical basis for the clinical immunotherapy of severe burn injuries.
