摘 要:目的探讨吸入式甲醛暴露对小鼠骨髓组织中转录因子GATA-2和CCAAT/增强子结合蛋白(C/EBPα)在mRNA以及蛋白表达水平的影响。方法将24只Balb/C小鼠随机分为3组,每组8只,采用仿真式口鼻暴露的方式,暴露于不同浓度的气态甲醛环境中(0、0.5和3.0 mg/m3),每天8 h,连续2周。染毒结束后,分别采用反转录PCR(RT-PCR)和酶联免疫吸附(ELISA)法测定小鼠骨髓组织中转录因子GATA-2和C/EBPαmRNA和蛋白的表达。结果与对照组相比,转录因子GATA-2和C/EBPαmRNA和蛋白的表达均随甲醛浓度的升高呈下降趋势,且甲醛浓度为3.0 mg/m3时,差异有统计学意义(P<0.05)。结论高浓度(3.0 mg/m3)甲醛暴露可以抑制小鼠骨髓组织中GATA-2和C/EBPαmRNA和蛋白的正常表达。Objective To investigate the impact of inhaled formaldehyde on the expression of transcription factor GATA-2 and C/EBPα at mRNA and protein level in mouse bone marrow. Methods A total of 24 Bale/C male mice were randomly divided into three groups, eight in each, and exposed to formaldehyde by nose-only inhalation at the doses of 0, 0.5, and 3.0 mg/m3 for 8 h/d, for two consecutive weeks. After two weeks of treatment, reverse transcription and polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) were used to detect the expressions of transcription factors GATA-2 and C/EBPα both at mRNA level and protein level. Results Compared with the control group, expression levels of GATA-2 and C/EBPα decreased significantly at the concentration of 3.0 mg/m3(p〈0.05); And with the increase of formaldehyde concentration, the expression levels showed a downward trend. Conclusion Formaldehyde exposure may affect the expression levels of GATA-2 and C/EBPα and cause damage to hemopoietic function of bone marrow at high exposure level.
