摘  要:目的探讨慢性铝暴露对大鼠海马组织中钙调蛋白(calmodulin,CaM)及cAMP反应元件结合蛋白(cAMPresponse element binding protein,CREB)mRNA和蛋白表达的影响。方法将40只初断乳清洁级Wistar雄性大鼠随机分为4组,分别为对照(蒸馏水)组和2、4、6 mg/ml三氯化铝染毒组,每组10只。采用自由饮水方式连续染毒3个月。检测大鼠脑组织中铝含量;采用Western blot方法检测大鼠海马组织中CaM和CREB蛋白的表达水平;采用RT-PCR方法检测海马组织中CaM和CREB mRNA的表达水平。结果与对照组相比,各浓度铝染毒组大鼠脑组织铝含量及CaM mRNA和蛋白的表达水平均升高,CREB mRNA和蛋白的表达水平均下降,差异有统计学意义(P<0.05)。随着铝染毒浓度的升高,大鼠脑组织铝含量及CaM mRNA和蛋白的表达水平均呈上升趋势,CREB mRNA和蛋白的表达水平均呈下降趋势。结论慢性铝暴露可促进大鼠海马神经元CaM mRNA和蛋白的表达,抑制CREB mRNA和蛋白的表达。Objective To investigate the effects of chronic aluminum(A1) exposure on calmodulin(CaM) and cAMP- response element binding protein (CREB) in hippocampus of rats. Methods Forty weaned Wistar rats were randomly divided into four groups, 10 rats in each group. The four groups were treated through drinking water at the doses of 0(control), 2,4 and 6 mg/ml A1C13 (A1 exposure) for three consecutive months,respectively. Graphite furnace atomic absorption spectrometry was used to determine A1 level in the brain. Western blot was used to detect the expression level of CaM and CREB in rat hippocampus. The mRNA expression levels were analyzed and quantified through reverse transcriptional PCR (RT-PCR). Results Compared with the control group, the protein and the mRNA expression levels of CaM and A1 levels in brain in A1- exposed groups were increased significantly (P〈0.05) ;The protein and the mRNA expression levels of CREB were decreased significantly (P〈0.05). With the increases of A1 doses the protein and the mRNA expression levels of CaM and A1 levels in the brain increased,the protein and the mRNA expression levels of CREB were decreased. Conclusion Chronic aluminum exposure may affect learning and memory through impacting on CaM and CREB expression in the hippocampus of rats.
