摘  要:为了鉴定牦牛乳脂肪滴形成和分泌相关的黄嘌呤脱氢酶(XDH)、嗜乳脂蛋白亚家族成员1(BTN1A1)基因全泌乳期的表达状况,产犊前15天和产犊后第1,15,30,60,120,240天采集5头麦洼牦牛乳腺组织样品,运用实时荧光定量PCR(RT-qPCR)技术分析XDH、BTN1A1基因全泌乳期表达变化以及它们与牦牛泌乳的关系。结果表明:牦牛乳腺BTN1A1基因在泌乳的第30天和第120天有2个表达峰,最大峰值出现在产犊后的第30天;XDH基因在泌乳的第30天出现峰值。XDH、BTN1A1基因在产犊后30~120 d的表达水平与产犊前15天相比差异显著或极显著(P<0.05或P<0.01),说明XDH、BTN1A1基因在牦牛产犊后30~120 d维持较高水平并稳定表达。To detect the expression patterns of xanthine dehydrogenase (XDH) and butyrophilin subfamily 1 member A1 ( BTNIA1 ) genes dur- ing the whole lactation, which are essential for lipid droplet formation and secretion, the mammary gland tissue samples from five Maiwa yaks were collected at 15 d before calving and 1 d, 15 d, 30 d, 60 d, 120 d and 240 d after calving. The expression patterns of the two genes, and their relationship with milk yield were detected using real - time quantitative PCR ( RT - qPCR). The results showed that two expression peaks were found in BTNIA1 gene at 30 d and 120 d of lactation, in which the high expression peak appeared at 30 d after calving. The high expres- sion peak of XDH gene appeared at 30 d after calving, and The expression levels of XDH and BTN1A1 genes from 30 d to 120 d after calving were significantly higher or extremely significantly higher than those at 15 d before calving ( P 〈 0.05 or P 〈 0.01 ). It indicated that the expres- sion levels of XDH and BTN1A1 genes were high and consistently.
