摘 要:目的:研究柴胡总皂甙对运动性疲劳大鼠海马、下丘脑单胺类神经递质及其代谢产物含量的影响。方法:将36只雄性Sd大鼠,随机均分为空白组、模型对照组和柴胡总皂甙组,共3组,每组12只。柴胡总皂甙组每天灌胃给药50mg/kg,模型对照组和空白对照组灌胃等量生理盐水,柴胡总皂甙组和模型组大鼠在灌胃1h后进行中等运动强度的水平跑台运动,速度为15m/min,坡度0°,跑台20min,间歇40min,再跑台20min,每天1次,连续14d。末次运动后处死大鼠并分离海马及下丘脑,HPLC-ECD法检测其单胺类神经递质及其代谢产物含量。结果:与空白组比较,模型组大鼠海马区5-HT、5-HIAA、NE含量升高,DA、DOPAC、MHPC含量降低;下丘脑区5-HT、5-HIAA含量升高,DA、DOPAC、NE、MHPC含量降低。与模型组比较,柴胡总皂甙组大鼠海马区5-HT、5-HIAA、NE含量降低,DA、DOPAC、MHPC含量升高;下丘脑区5-HT、5-HIAA含量降低,DA、DOPAC、MPHC含量升高。结论:柴胡总皂甙抗运动性疲劳,可改善运动性疲劳中枢单胺类神经递质合成与代谢紊乱的状况,可作为主要成分广泛应用于抗疲劳食品和药品的市场开发中。Objective: To study the effect of saikosaponin on central monoamine neurotransmitters and contents of monoamine in sport fatigue rats. Method: 36 SD male rats were divided into 3 groups with 12 rats in each group randomly: blank group, model group and saikosaponin group. The saikosaponin group was given gastric garage of saikosaponin once 50 mg/kg a day, while rats in blank group and model group were given the same volume of normal saline. One hour after administration, the saikosaponin group and model group received medium-intensity treadmill exercise for 20 minutes at the speed of 15 m/min, with a slope of 0 degree, and after 40 minute break repeated it for another 20 minutes. Fourteen days later, killed these rats and get the hippocampus and hypothalamus of them after the last treadmill training. Then test its monoamine neurotransmitters and contents of monoamine by the method of HPLC- ECD. Results: Compare to the blank group, the content of 5-HT, 5-HIAA, NE in hippocampus of the model group rats increased, the content of DA, DOPAC, MHPC in hippocampus reduced; the content of 5-HT, 5-HIAA in hypothalamus of the model group rats increased, the content of DA, DOPAC, NE and MHPC in hypothalamus reduced. Compare to the model group, the content of 5-HT, 5-HIAA, NE in hippocampus of the saikosaponin group rats reduced, the content of DA, DOPAC, MHPC in hippocampus increased; the content of 5-HT, 5-HIAA in hypothalamus of the saikosaponin group rats reduced, the content of DA, DOPAC and MHPC in hypothalamus increased. Conclusion: The anti-fatigue effect of saikosaponin is involved in improving the synthesis of central monoamine neurotransmitters and the condition of metabolic disorder. Thus saikosaponin can be widely used in the market development of anti-fatigue food and medicine as their primary components.
