摘 要:为研究库拉索芦荟皮多糖提取的工艺条件,以蒽酮-浓硫酸法测定粗多糖中的总糖含量为指标,采用正交实验L9(33)结合加速溶剂萃取技术(ASE)对库拉索芦荟皮多糖提取工艺进行优化设计。结果显示,库拉索芦荟皮多糖的最佳提取工艺条件为温度180℃,静态提取3次,每次18min,提取温度对多糖提取率具有极显著性影响。在最优条件下,库拉索芦荟皮多糖提取率为(9.03±0.12)%。与传统水提醇沉提取法(多糖提取率6.9%)相比,该提取工艺具有操作简便、缩短提取时间、节能耗、多糖得率高等优点,是一种快速经济实用的新型工艺。To study the extraction conditions of polysaccharide from skin of Aloe vero, the total content of the crude polysaccharide, as a indicator, was determined by anthrone-sulfuric acid method, and the extraction technology of the polysaccharide from skin of Aloe vero was optimized with L9(33) orthogonal experimental design and accelerated solvent extraction (ASE) process. Results showed that the optimum extraction conditions were 180 ℃, 3 times with 18 min each time. The extraction temperature was a significant factor. Under the optimal conditions, the yield of polysaccharide from skin of Aloe vero reached as high as (9.03±0.12)%. Compared with the traditional extraction technology of water extraction and alcohol precipitation (6.9% extraction yield), the extraction technology in this paper is simple, rapid, energv-saving and efficient, it's a fast. economical, practical and new technology.
