诊断超声照射后人早孕绒毛Fas/FasL蛋白表达的研究

摘　　要：目的 :检测诊断超声辐照与人早孕绒毛 Fas/ Fas L蛋白表达的关系。方法 :对拟进行人工流产的 2 4例早孕 (4 5～ 6 0 d)妇女随机分为 4组 :对照组为空白照射组、10 min组、2 0 min组和 30 min组。应用 Hp85 0 0彩色多普勒超声诊断仪 ,经腹对孕囊进行不同时间的持续照射 ,照射后 2 4h取材。用免疫组织化学技术检测上述各组绒毛滋养层细胞 Fas/ Fas L 蛋白表达情况。结果 :超声照射后 10 min组绒毛滋养层细胞 Fas/ Fas L蛋白表达率与对照组无差别 (P>0 .0 5 ) ;2 0 min组和 30 min组滋养层细胞表达率明显增加 ,显著大于对照组和 10 min组 (P<0 .0 0 1)。结论 :诊断超声持续照射早孕孕囊组织 >10 min可引起绒毛滋养层细胞 Fas/ Fas L 蛋白表达率增加 。Objective:To investigate the role of Fas/FasL protein expression in first trimester pregnancy trophoblast after diagnostic ultrasound irradiation.Methods:Twenty four first trimester pregnancy women were randomly separated into egual four groups according to different irradiating time:control group(bland radiation),10 min group, 20 min group and 30 min group. Gestational sac were irradiated by HP 8500 color Doppler ultrasonography with 7 5MHz linear phased array transducer. All the trophoblasts were removed at 24 h after. Expression of Fas/FasL protein was analyzed with immunohistochemistry.Results:The positive rates of Fas/FasL protein in experiment groups 10 min group excluded were all higher. The rates of expression of Fas and FasL in control group trophoblasts were (32 56±5 23)%,(46 44±4 39)% respectively. After irradiation the positive rate was increased in 20 min group and 30min group,Fas expression peaking to(45 63±9 38)% in group 30min trophoblasts;FasL expression peaking to (54 05±8 67)% in group 30 min trophoblasts. There was no difference between control group and 10 min group ( P >0 05). Conclusion:Continuous irradiation over 10 minutes by diagnostic ultrasound may result in higher expression of Fas and FasL in first trimester prognancy, further inducing trophoblasts cell apoptosis resulted.
