摘  要:目的从酵母菌中提取其多糖成分,探讨酵母多糖对小鼠免疫功能的影响,为酵母多糖的临床应用提供实验依据。方法取小鼠脾脏淋巴细胞,置于96孔细胞培养板中,加入酵母多糖为酵母多糖组和细胞培养液为空白对照组,测定小鼠脾细胞的增殖活性;取细胞上清液,测定细胞因子白细胞介素-2(IL-2)、干扰素-γ(IFN-γ);用流式细胞术(FCM)检测细胞表型,观察酵母多糖对细胞免疫和体液免疫作用。结果实验组细胞增殖活性显著高于对照组(P<0.01);实验组IL-2,IFN-γ的浓度与酵母多糖在一定浓度范围内成剂量依赖关系;酵母多糖即能刺激B淋巴细胞,也能刺激T细增殖。结论酵母多糖可提高小鼠免疫功能,它不仅能提高体液免疫功能,而且能提高细胞免疫功能。Objective To investigate the effects of yeast polysaccharides (YPS) on immune function in order to provide experimental basis for clinical application. Methods The spleen cells of mice were taken and placed in 96-well plate,which were divided into two groups,YPS group with YPS and control group without YPS. The proliferation activities of the cells were detected by WST-1 assay; the concentrations of IL-2, IFN-γ in supernatant were examined by ELISA;the cell phenotypes were detected by flow cytometry (FCM). Results As compared with those in control gronp: the plo!iferation activities of the cells in YPS group were significantly increased( P 〈0.01). There was a dose-dependent relationship between concentrations of IL-2, IFN-γ and YPS in YPS group. Moreover, YPS could stimulate the proliferation both T ceils and B ceils. Conclusion YPS can enhance immune function of mice, moreover, which can not only increase humoral immune function, but also increase cellular immune function.
