扶正解毒方对肺腺癌A549细胞凋亡及凋亡基因Survivin表达的影响

摘　　要：目的研究扶正解毒方对肺腺癌A549细胞凋亡及凋亡基因Survivin表达的影响,探讨该方可能具有的抗癌机制。方法应用血清药理学方法,对A549细胞株进行体外培养,予不同剂量扶正解毒方及0.9%氯化钠溶液含药血清进行干预,流式细胞术检测各组细胞凋亡率,免疫组织化学法检测凋亡基因Survivin表达。结果扶正解毒方各剂量组平均凋亡率均高于0.9%氯化钠溶液组凋亡率,各组差异均具有统计学意义(P<0.01,P<0.05)。免疫组织化学法结果显示,扶正解毒方各剂量组Survivin蛋白表达较生理盐水组降低,其中,扶正解毒方高、中剂量组与对照组相比差异具有统计学意义(P<0.01,P<0.05)。结论扶正解毒方能促进肿瘤细胞凋亡,抑制凋亡基因Survivin的表达。Objective To study the effects of Fuzheng Jiedu Recipe （FJR） on A549 cell apoptosis and the expression of survivin gene, and explore the probably anti-tumor mechanism. Methods With the serologic pharmacological method, lung adenocarcinoma A549 cells was treated by FJR and apoptosis of A549 ceils and survivin expression were detected by flow cytometry and immunohistochemistry respectively. Results The average apoptosis rate in each FJR group was higher than that in control group（P〈0. 01, P〈 0. 05）. The level of survivin protein in each FJR group was decreased, compared with that in control group（P〈0. 01, P〈0. 05 ）. Conclusion FJR could induce lung adenocarcinoma A549 cells apoptosis and inhibit the expression of survivin protein.
