摘  要:目的通过对H_(22)荷瘤小鼠肿瘤组织基因表达谱的检测,探讨消癌解毒方抗肿瘤的作用机制。方法 H_(22)荷瘤小鼠随机分为空白对照组,消癌解毒方低、中、高剂量组和顺铂组。应用基因芯片技术,检测各组H_(22)荷瘤小鼠肿瘤组织的差异表达基因;应用pathway分析,找出与消癌解毒方抗癌机制相关的信号通路和基因,并针对趋化因子信号通路进行分析。结果消癌解毒方能显著影响多个与肿瘤生长、凋亡和免疫相关的信号通路,并能下调趋化因子信号通路中CCL3、CXCL2等基因的表达。结论消癌解毒方可能通过影响趋化因子信号通路中的某些基因的表达来发挥抗肿瘤生长和侵袭的作用。Objective To explore the mechanism of Xiaoai Jiedu Recipe (X JR) for fighting against tumors by detecting tumor gene expression profiles of H22 tumor-bearing mice. Methods H22 tumor-bear- ing mice were randomly divided into the normal control group, the low dose X JR group, the medium dose X JR group, the high dose X JR group, and the Cisplatin group. The differentially expressed genes of tumor tissues in H22 tumor-bearing mice were detected by using gene chip technique. The antitumor mechanism of X JR associated signaling pathways and gene expressions were found out by pathway analysis. The chemo- kine signaling pathways were analyzed. Results XJP could significantly affect multiple signaling pathways associated with tumor growth, apoptosis, and immunity. XJP also could decrease expressions of CCL3 and CXCL2 in the chemokine signaling pathway. Conclusion XJP could inhibit the growth and invasion of tumor cells possibly by affecting expressions of some genes in the chemokine signaling pathway.
