摘 要:目的探讨B细胞活化因子(B cell-activating factor,BAFF)在伴和不伴哮喘的鼻息肉组织中的表达情况。方法 18例鼻息肉患者(其中8例合并哮喘)和10例正常对照者,分别采用免疫组化(immunohistochemistry,IHC)、定量逆转录聚合酶链反应(quantitative reverse transcription polymerase chainreaction,qRT-PCR)检测鼻息肉和对照鼻甲组织中BAFF的定位、mRNA表达情况,并比较伴和不伴哮喘的鼻息肉组织中BAFF mRNA和蛋白表达的差异。结果鼻息肉组织中BAFF存在广泛阳性染色,而对照组织为弱阳性染色,2组阳性细胞数分别为4.05[2.01,6.72]/HPF和1.74[0.68,2.47]/HPF。鼻息肉组织中BAFF mRNA的表达分别为5.30[2.42,7.92]和1.20[0.51,1.95]),差异有显著性意义(P<0.05)。两个亚组进行比较,BAFF mRNA和蛋白表达在伴随哮喘的鼻息肉组织中较不伴随哮喘的嗜酸性鼻息肉组织中显著增高,mRNA:8.35[6.35,14.2]和2.85[1.20,4.47];蛋白:424.8[332.3,475.8]和276.3[161.0,322.6],差异有显著性意义(P<0.05)。结论鼻息肉组织中BAFF的表达增高可能是与鼻息肉伴发哮喘与否存在关联性。OBJECTIVE To examine the expression of B cell-activating factor (BAFF) in nasal polyps (NP) with or without concomitant asthma. METHODS Eighteen adult NP patients (asthmatics=8) and 10 normal controls were enrolled to investigate the level of BAFF in polyp tissues by using immunohistochemistric staining, quantitative reverse transcription polymerase chain reaction (qRT- PCR) and ELISA. RESULTS Compared with the normal control, the BAFF immunoreactivity and mRNA level were significantly upregulated in polyp tissues (number of positive cells, 4.0512.01, 6.72]/HPF vs 1.7410.68, 2.47]/HPF) ;mRNA level, 5.30[2.42, 7.92] vs 1.20[0.51, 1.95]) (P〈0.05) . When subdivided NP patients into asthmatic and non-asthmatic subgroups, we found BAFF mRNA and protein level were significantly higher in asthmatic subgroup than in non-asthmatic subgroup (mRNA, 8.35[6.35, 14.2] vs 2.8511.20, 4.47]; protein, 424.8[332.3, 475.8] vs 276.31161.0, 322.6]) (P〈0.05) . CONCLUSION Our findings suggested BAFF upregulation in NP tissues may be associated with concomitant asthma.
