摘  要:目的观察小鼠自体骨髓单个核细胞(BMMNC)移植后有无输注不良反应,是否形成肝内及肝外血栓及宿主凝血机制有无其他异常表现。方法应用CCl4/2-AAF制备小鼠急性肝损伤模型,然后行BMMNC移植。实验按注射BMMNC的数量随机分成3组:A组为移植对照组;B组为经尾静脉注射BMMNC0.3ml;C组为经尾静脉注射BMMNC0.9ml;分别于移植后1周、2周处死小鼠,取其血清检测D-二聚体值,取肝、肺、心、脑、肾组织观察小鼠BMMNC移植后这些器官有无血栓形成。结果可在肝脏损伤处发现红色荧光;同一时间点各实验组间比较、A组与其他组比较、同一实验组移植后1周与2周比较均有统计学差异(P<0.05)。各器官血栓形成情况:HE染色观察发现,1周及2周肝脏、心脏、肺脏及肾脏均有血栓形成,第2周血栓较第1周明显,脑部未发现血栓。D-二聚体检测:乳胶凝集法检测血清D-二聚体值,各时间点各组间均未发现差异。结论 BALB/C小鼠尾静脉注射BMMNC0.3ml及0.9ml均可致多器官血栓形成。BALB/C小鼠尾静脉注入BMMNC剂量大者形成血栓的机会越大。乳胶凝集法测定D-二聚体值对检测BALB/C小鼠尾静脉移植BMMNC后是否有血栓形成没有帮助。Objective To observe the adverse reaction, the formation of intrahepatic and extrahepatic thrombosis and the host coagulation system other unusual behavior after autologous bone marrow mononuclear cell transplantation. Methods Acute liver failure model was made by CCl4/2-AAF,then, BMMCs were infused into the caudal veins of dif- ferent mice. All mice were divided into 3 groups randomly: A: control treatment; B:0.3 ml BMMNC; C:0.9 ml BMMNC. 1 week and 2 weeks after transplantation, the values of plasma D-dimer were assessed, the livers, kidneys, lungs, hearts and brains were examined anatomically, and the thrombus found in these organs were further examined pathologically. Results Fluorescent was found in livers. Histological finding: thrombosis could be found in the hearts, the livers, the lungs and the kidneys; Latex agglutination detection of serum D-dimer values, there was not different a- mong three groups at each time point. Conclusion Thrombosis can be found in multiple organs in BALE/C mice by tail vein injection of 0.3 ml and 0.9 ml BMMNC.
