摘 要:目的 探讨乙醛脱氢酶2(ALDH2)基因多态性与男性酒依赖综合征的关系.方法 应用聚合酶链式反应结合限制性片段长度多态性(PCR-RFLP)分析法,对ALDH2基因的特定片段进行特异性扩增,限制性内切酶酶切,非变性聚丙烯酰胺凝胶垂直电泳,结合硝酸银染色法判定基因型.结果 病例组ALDH2 1*1(G/G),ALDH2 1*2(G/L)和ALDH2 2*2(L/L)基因型的分布频度分别为53.68%,38.42%和7.89%,对照组分别为41.89%,49.10%和9.01%,两组之间的基因型频度分布差异无统计学显著性意义(χ2=5.786,P=0.055).与携带ALDH2 1*1(G/G)基因型者相比,携带ALDH2 1*2(G/L)和ALDH2 2*2(L/L)基因型者的比值比(odds ratio,OR)都有所降低.与携带ALDH 2*2等位基因者相比,ALDH2 1*1(G/G)基因型者发生酒依赖综合征的风险显著升高,其OR为1.79,95%可信限(95% confidence interval,95%CI)为1.19~2.69.结论 酒依赖综合征患者组与健康对照组之间ALDH2的基因型频度分布差异无显著性.与携带ALDH2 1*1(G/G)基因型者相比,携ALDH2 1*2(G/L)和ALDH2 2*2(L/L)基因型者的OR值都有所降低.与携带ALDH2 2*2等位基因者相比,ALDH2 1*1(G/G)基因型者发生酒依赖综合征的风险显著升高.Objective To study the relationshiop with the acetaldehyde dehydrogenase 2 (ALDH2) gene polymorphism and male patients of alcohol dependence syndrome. Methods Sample of vein blood combined the EDTA, extracted deoxyribonu- cleotide(DNA) ,adopted with polymerase chain reaction (PCR) combining with Restriction-Fragment-length polymorphism (RFLP) ,the genotype of the ALDH2 were detected by polyacrylamide gel electrphoresis (PAGE). Results The gene fre- quency of the ALDH2 1 * 1 (G/G),ALDH2 1 *2 (G/L) and ALDH2 2 * 2 (L/L) were 53.68%,38. 42% and 7.89% in the patients class, gene frequency of the two classes distributes no significant difference (x2 5. 786, P = 0. 055). And the odds ratio(OR) of the ALDH2 1 * 2 (G/L) and ALDH2 2 * 2 (L/L) genotypes was lower,compared to ALDH2 1 * 1 (G/ G) genotypes. And the OR of the ALDH2 1 * 1 (G/G) was higher than ALDH2 2 ~ 2,and its OR was 1.79,95% confidence interval (95~CI) was 1.19~2.69. Conclusion 180 patients of alcohol dependence syndrome and 240 health collators,the gene frequency of the two classes of the ALDH2 distributes had no significant difference. The odds ratio (OR) of the AL- DH2 1 * 2 (G/L)and ALDH2 2 ~ 2 (L/L) genotypes was lower than ALDH2 1 ~ 1 (G/G) genotypes. The odds ratio (OR) of the ALDH2 1 * 1 (G/G) was higher,compared to ALDH2 2 * 2. The risk for alcohol dependence syndrome with ALDH2 1 * 1 (G/G) was higher than the person with ALDH2 2 * 2. It is said that ALDH 2 * 2 allele may be a decreased risk factor of alcohol dependence syndrome.
