摘 要:目的 分析5种致病性弧菌在不同NaCl浓度碱性蛋白胨水增菌液中的生长情况,筛选共增菌的最适NaCl浓度.方法 选择副溶血性弧菌、非O1非O139群霍乱弧菌、创伤弧菌、溶藻弧菌、拟态弧菌作为研究菌株,分别在含5,10,20和30 g/L NaCl的碱性蛋白胨水中36℃培养24 h,每隔1 h连续测定波长490 nm的吸光度值A,绘制生长曲线,分析不同NaCl浓度对5种致病性弧菌生长的影响.结果 不同NaCl浓度对5种致病性弧菌生长的影响不同.副溶血性弧菌、溶藻弧菌、非O1非O139群霍乱弧菌的增殖能力优于创伤弧菌、拟态弧菌.NaCl浓度升高,溶藻弧菌的迟缓期不变,而其余4株致病性弧菌的迟缓期都出现不同程度的延长;NaCl浓度升高,副溶血性弧菌的增殖能力不受影响,溶藻弧菌的增殖能力出现增强趋势,10,20和30 g/L的NaCl浓度更有利于生长,而其余3株致病性弧菌的增殖能力出现减弱的趋势,5和10 g/L的NaCl浓度更有利于生长.结论 10 g/L为最适NaCl浓度,含10 g/L NaCl的碱性蛋白胨水增菌液可以满足同时检测5种致病性弧菌的增菌需要.Objective To investigate the growth of five strains pathogenic vibrio in alkaline peptone water enrichment broth at different NaCl concentration and select the best NaCl concentration in order to enrich these strains simultaneously. Methods Five strains pathogenic Vibrio,including Vibrio parahaemolyticus, non-O1 and non-O139 Vibrio cholerae ,Vibrio vulnifi- cus ,Vibrio alginolyticus, and Vibrio mimicus, were cultured in the alkaline peptone water enrichment broth at different NaCI concentration (5,10,20,30 g/L) for 24 hours at 36℃. Every one hour,the value of A 490nm of the culture was detected. And its impacts on the growth of these pathogenic Vibrio were observed by growth curve. Result Different NaCl concentra- tion had different effects to 5 strains pathogenic Vibrio. The proliferation ability of three strains (V. parahaemolyticus,V. alginolyticus,non-O1 and non-O139 V. cholerae)was better than that of the two strains (V. vulni ficus,V, mirnicus). The lag phase of V. alginolyticus remained unchanged when the NaC1 concentration in broth were increased,but that of the oth- ers were delayed in different degree. The proliferation ability of V. parahaemolyticus remained unchanged when the NaCl concentration in broth were increased ,but that of V. alginolyticus was increased and those of the other three strains were re- duced. The NaCl concentration (10,20,30 g/L) were better for the growth of V. alginolyticus,but the NaCl concentration (Sg/L and 10 g/L) were better for the growth of the other three strains (non-01 and non-O139 V. cholerae,V, mimicus and V. vulnificus). Conclusion 10 g/L was the best NaC1 concentration. Alkaline peptone water enrichment broth with 10 g/L NaCl could satisfy the request for enriching these five stains simultaneously.
