摘 要:目的观察培土生金法对慢性阻塞性肺疾病(COPD)稳定期模型大鼠肺组织线粒体膜电位(△·ψm)、肺组织病理的影响。方法 120只SD大鼠随机分成正常对照组(n=20)和模型组(n=100)。模型组采用气管内注射脂多糖加烟熏的方法,制作COPD大鼠模型,从模型组中选出大鼠80只,再随机分为黄芪建中汤(高、中、低剂量组)、金水宝胶囊组和生理盐水组。给药8周后,用流式细胞仪检测肺组织线粒体膜电位,在光镜下观察肺病理形态变化。结果 COPD模型组大鼠见肺气肿病理形态变化,黄芪建中汤可明显改善COPD模型组支气管壁和肺泡炎性细胞浸润、肺泡扩张融合等病理改变;COPD模型组大鼠肺脏组织中线粒体膜电位较正常对照组明显下降(P<0.01);与模型组相比:黄芪建中汤(高、中、低剂量组)、金水宝胶囊组肺脏组织中线粒体膜电位明显升高(P<0.01),升高的次序为黄芪建中汤高剂量组>黄芪建中汤中剂量组>金水宝胶囊组>黄芪建中汤低剂量组。黄芪建中汤高剂量组与黄芪建中汤中剂量组、黄芪建中汤中剂量组与金水宝胶囊组,金水宝胶囊组与黄芪建中汤低剂量组之间差异均有统计学意义(P<0.01)。结论通过培土生金法治疗,不仅对COPD大鼠呼吸系统的症状和体征有明显改善,而且可以显著提高肺组织线粒体膜电位,对COPD稳定期的防治具有一定的理论和实践意义。Objective To observe the prevention and treatment of the method of strengthening earth to generate metal on rat lung tissue mitochondrial membrane potential in the stabilization period of chronic obstructive pulmonary disease, and its influence on the lung tissue pathology. Methods 120 SD rats were randomly divided into normal control group (n = 20) and model group ( n = 100). In model group rat model of COPD was made by endotracheal injection of lipopolysaccharide(LPS) plus smoked method. Among the rats 80 were selected and randomly divided into Huangqi Jianzhong Decoction (high, middle, low) dose group, Jinshui Bao capsule group and normal saline group. The mitoehondrial membrane potential was detected 8 weeks after the treatment by flow cytometry in lung tissue, and pulmonary pathological changes were observed under light microscope. Results Emphysema pathological morphological changes could be seen in COPD model group rats. Compared with COPD model group, bronchial wall and alveolar infiltration of inflammatory cells, alveolar expansion of integration and other pathological changes in the treatment group showed obvious improvement, and the most obvious improvement could be seen in Huangqi Jianzhong Decoction high dose group. The mitochondrial membrane potential in lung tissue of rats in COPD model group decreased significantly than that of the normal control group(P 〈 0.01 ) ; Compared with model group, mitochondrial membrane potential in lung tissue of treatment group was significantly increased (P 〈 0.01 ), and in terms of the increasing effect, Huangqi Jianzhong Decoction high dose group was better than middle dose group, Huangqi Jianzhong Decoction middle dose group was better than Jinshui Bao capsule group, and Jinshui Bao capsule group was better than Huangqi Jianzhong Decoction low dose group.
