RP-HPLC法测定地黄及其复方制剂中尿苷和腺苷的含量

摘　　要：目的分别建立HPLC外标法测定地黄药材及其制剂中尿苷和腺苷含量的方法。方法采用Thermo C18柱(250 mm×4.6 mm,5μm),尿苷以5 mmol·L-1醋酸铵(乙酸调节pH为6.0)为流动相,腺苷以20 mmol·L-1磷酸二氢钾∶甲醇(90∶10)为流动相,流速均为1 mL·min-1,检测波长均为254 nm。结果尿苷浓度在4.17～20.85 mg·L-1(r=0.999 9)范围内线性关系良好,腺苷浓度在1.01～20.23 mg·L-1(r=0.999 8)范围内线性关系良好,平均加样回收率(n=9)分别为100.8%和100.1%。测得6批地黄药材中尿苷的平均含量为(0.027 8±0.003 5)%,腺苷的平均含量为(0.008 4±0.003 1)%。制剂中尿苷含量为0.032 1%,腺苷含量为0.006 9%。结论建立的方法准确可靠,专属性好,可作为地黄及其制剂中核苷含量测定方法。地黄药材中尿苷是主要的核苷成分,其它核苷成分含量甚微。Objective To develop an HPLC method for determination of uridine and adenosine in Radix Rehmanniae and Danzhi Jiang-tang Capsules. Methods The chromatographic separation was achieved on a Thermo Cls column （250 mm×4.6 mm,5 μm） with 5 mmol·L^-1 ammonium acetate and 20 mmol·L^-1 potassium dihydrogen phosphate-methanol （90：10） as mobile phases for uridine and adenosine,respectively,at a flow rate of I mg·L^-1 and UV detection at 254nm. Results The linear range for uridine was 4.17 - 20.85 mg ·L^-1（ r = 0. 999 9） and for adenosine 1.011 - 20.23 mg ·L^-1（ r = 0. 999 9）. The average recoveries （ n = 9 ） of uridine and adenosine were 100.8% and 100. 1 % , respectively. The uridine and adenosine contents found in 6 batches of Radix Rehmanniae and Prepared Radix Rehmanniae were （0.027 8 ±0.003 5） % and （0.008 4±0.003 1 ） % ,respectively. The uridine and adenosine contents found in Danzhi Jiangtang Capsule were 0. 032 1% and 0.006 9% , respectively. Conclusion The established methods are selective, re-liable and applicable for the quality control of nucleoside in Radix Rehmanniae and Danzhi Jiangtang Capsules, and uridine is found to be the main component with others in very minute contents.
