摘  要:目的 探讨胎儿卵巢能否用于临床移植。方法 取自流产或病理原因水囊引产的胎儿卵巢共 49份。将卵巢切成碎块进行组织培养 ,培养后观察组织学变化 ,同时测定培养液中雌二醇 (E2 )浓度 ;取培养前后胎儿卵巢用免疫组化法检测FSH -Binding、LH -Binding、LH -R、HLA -ABC、HLA -DR抗原含量 ;观察冷冻保存后卵泡形态发育及E2 分泌。结果 胎儿卵巢在体外培养条件下 ,能进一步发育 ,而且能分泌E2 ;胎儿卵巢最早于 2 0周出现FSH -Binding、LH -Binding、LH -R阳性表达 ;胎儿卵巢HLA -ABC、HLA -DR抗原含量明显低于成人卵巢 ,经体外培养后HLA -DR抗原含量进一步降低 ;胎儿卵巢复苏培养后卵泡仍能进一步发育和产生E2 。结论 胎儿卵巢可用于卵巢移植 。Objective\ To study whether the fetal ovary could be used for transplantation to patients with premature ovary failure and bilateral removal of ovary.Methods\ 49 fetal ovaries were obtained from fetuses of 17 to 27 gestational weaks.The fetal ovaries were cultured and the ovarian tissue were fixed for histological examination after culturing for 3,6 and 9 days.The estrodiol concentration in the culture medium were determined.The appearance time of FSH-Binding,LH-Binding and LH receptor in 32 human fetal ovaries were investigated by LSAB immunohistochemistry.The antigen contents of HLA-ABC and HLA-DR were detected by ABC immunohistochemistry.Ovaries for cryopreservation were suspended in 10% dimethyl sulfoxide RPMI 1640 solution and put into liquid nitrogen by threes ctep methods,and the fast thawing method was used.The frozen-thawed tissue was examined with the same methods as above.Results\ The fetal follicles could develop to more mature stages and secrete estrodial in tissue culture and the amount of estrodial increased with the long culture time.The positive expression of FSH-Binding,LH-Binding and LH-R were detected only in tissue obtained above 20 gestational weeks.The antigen contents of HLA-ABC and HLA-DR in fetal ovaries were significantly lower than those in adult ovaries.The HLA-DR content of fetal ovaries decreased after culturing in vitro.The follicles in frozen-thawed ovaries could further develop and secrete estrodial after culture and these were similar to that in non-cryopreserved group.Conclustion\ The human fetal ovary could be used for clinical ovary transplantation and seemed to be superior to adult ovary.
