摘 要:目的观察芹菜素(Apigenin)对小鼠睾丸组织抗氧化酶活性及抗氧化能力的影响,探讨AP对雄性小鼠生殖功能影响机制。方法 200只成年健康SPF级昆明种雄性小鼠按体重随机分为4组:对照组(生理盐水10ml/kg)、低剂量组(AP 252mg/kg)、中剂量组(AP 504mg/kg)和高剂量组(AP 1008mg/kg),每组50只。连续灌胃7、14、21、28、35天,分批处死,取出睾丸,制备匀浆,测定总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活力及酰胺腺嘌呤二核苷酸磷酸(NADPH)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)的浓度。结果 7天CAT活力低、中、高剂量组均高于对照组,其中低、中剂量组与对照组差异有统计学意义(P<0.05)。GSH-PX活力,T-SOD活力、MDA浓度及GSH/GSSG在不同时间观察点,各剂量组与同期对照组差异均无统计学意义。T-SOD活力随时间变化总体升高,MDA浓度随时间变化总体降低,GSH/GSSG高剂量组在不同时间观察点均降低。结论本实验条件下,AP可致小鼠睾丸组织呈现氧化应激状态,对小鼠睾丸具有一定的亚急性氧化损伤作用。Objective: Observe the influence of apigenin on the activity of antioxidant enzymes in testis of mice, in order to explore the influence mechanism of AP to reproductive function in mice. Methods : 200 healthy adult male mice of Kunming strain in SPF degree were randomly divided into four groups ( 50 mice in every group) : control group ( 10ml/kg NS), low - dose group ( AP 252mg/kg), middle dose group ( AP 504mg/kg) and high dose groups ( AP 1008mg/kg). The mice had been given NS or AP daily by intragastric administration. The mice of every group were killed at each time interval (7, 14, 21,28, 35d) respectively. To determine the activity of superoxide dismutase ( SOD), catalase ( CAT), glutathione peroxidase ( GSH - PX) and nicotinamide adenine dinucleotide phosphate hydride ( NADPH), the concentration of malondiadehyde ( MDA), the total antioxidative capacity ( T - AOC), glutathione (GSH) and oxidized glutathione (GSSG). Results: The activity of CAT increased in the low, middle and high dose group at 7 days compared with the control group, the difference was statistical between the low and middle dose group (P 〈 0. 05 ). Compared with the control group, the differences of GSH - PX, T - SOD, MDA and GSI-I/GSSG' activity in each dose group were not statistically significant. The activity of T - SOD generally incresed and the concentration of MDA generally reduced with time changing. GSH/GSSG in high dose group reduced in different observation point. Conclusion: In this experiment condition, AP could cause oxidative stress state and some subacute oxidative damage effect to testicular tissue in mice.
