摘 要:目的探讨血管紧张素Ⅱ(AngⅡ)及其受体AT1在心肌肥厚中的作用,研究DMP811(AT1受体拮抗剂)、培哚普利(ACEI)对AngⅡ诱导的心肌肥厚大鼠心肌AT1mRNA表达水平的影响。方法 24只6周龄的SD雄性大鼠随机分为4组:正常对照组、AngⅡ组、AngⅡ+DMP811组、AngⅡ+培哚普利组,每组6只。分别皮下埋藏装有生理盐水或AngⅡ的渗透微泵。1周后观察心脏质量、心脏质量/体质量的变化,采用反转录聚合酶链反应(RT-PCR)检测心肌AT1mRNA表达情况,并进行比较。结果①输注AngⅡ7 d不会明显影响大鼠的体质量,但可使大鼠的心脏质量、心脏质量/体质量显著增加。AngⅡ组较正常对照组心脏质量、心脏质量/体质重明显增加(P<0.05);与AngⅡ+DMP811组、AngⅡ+培哚普利组相比,AngⅡ组心脏质量、心脏质量/体质量也明显增加,差异具有统计学意义(P<0.05),而AngⅡ+DMP811组、AngⅡ+培哚普利组的心脏质量、心脏质量/体质量差异无统计学意义(P>0.05)。②AngⅡ组的AT1mRNA水平较对照组上调(84.5±3.0)%(P<0.01),DMP811、培哚普利药物干预,可使AngⅡ诱导的AT1mRNA的转录上调有效阻断。而AngⅡ+DMP811组、AngⅡ+培哚普利组的AT1mRNA水平差异无统计学意义(P>0.05)。结论心肌肥厚时AT1mRNA的表达水平明显增加,且AngⅡ诱导了AT1mRNA的转录上调。而AngⅡ导致的上述改变能被DMP811、培哚普利有效阻断,为以后防治心肌肥厚提供了重要依据。Objective To explore the effect of Ang Ⅱ and its receptor AT, with myocardial hypertrophy, study the im- pact of DMP811 (A% antagon) and perindopril (ACEI) on the expression of AT1 gene in cardiac muscle of myocardial hypertrophy rats induced by Ang Ⅱ .Methods 24 six-week old SD male rats were randomized to 4 groups which in- eluded control group,Ang Ⅱ group,Ang Ⅱ plus DMP811 group,Ang 11 plus perindopril group,they were respectively im- planted controlled porosity osmotic pumps containing normal saline or Ang Ⅱ subcutaneously. The change of heart weight, the ratio of heart weight to body weight was observed after 7 days treatment. Reverse transcription-polymerase chain reaction (RT-PCR) was used to check the expression of A% mRNA, and data was analyzed. Results (1)Infusion of Ang 11 for 7 days, there was no remarkable impact on rats' weight, however, the heart weight, ratio of heart weight to body weight of rats were dramatically increased when compared with control group. And the differences were signifi- cant(P〈O.05).The heart weight and ratio of heart weight to body weight increased remarkably in Ang 11 group compared with control group (P〈0.05); when compared with Ang 11 plus DMP811 group and Ang Ⅱ plus perindopril group, the heart weight and ratio of heart weight to body weight in Ang 11 group were also increased obviously(P〈0.05). However, there was no significant differences of heart weight and ratio of heart weight to body weight between Ang 11 plus DMP811 group and Ang Ⅱ plus perindopril group (P〉0.05).(2)The expression level of AT, mRNA increases by (84.5± 3.0)% (P〈0.01) in Ang 1t group compared with control group.DMP811,perindopril can inhibit the transcriptional of ATlmRNA induced by Ang Ⅱ effectively. But there were no significant differences of A% mRNA expression level be- tween Ang Ⅱ plus DMP811 group and Ang Ⅱ plus perindopril group (P〉0.05).Conchlsion Angiotensin Ⅱ could induce the expression of angiotensin 11-1
