蓖麻毒素气源性中毒对小鼠免疫屏障的影响

摘  要:蓖麻毒素来源广、毒性强、性质稳定,可通过注射、口服、气溶胶等多种方式中毒。为进一步优化蓖麻毒素气溶胶中毒模型,探讨蓖麻毒素气溶胶粒子对气—血屏障、血—胸屏障等免疫屏障的损伤过程,以及实质性免疫器官的病理改变,利用军事医学科学院军事兽医研究所五室已经建立的气溶胶暴露系统,对小鼠进行蓖麻毒素气溶胶的暴露,并于气溶胶暴露后12,30,48 h采集小鼠气管、肺脏、胸腺、脾脏制作病理切片,观察病理变化。结果表明:蓖麻毒素气溶胶粒子能够引起间质性肺炎、脾脏及胸腺细胞变性坏死,从而验证吸入蓖麻毒素气溶胶粒子对免疫屏障的强烈损伤。Ricin is of wide source,strong toxicity and stable nature,and can be exposed to various ways like injection,oral taking and aerosol.In order to further optimize the model of ricin aerosol exposure,explore the injury process of ricin aerosol exposure aerosol-blood barrier,blood-thymus barriers and other immune barriers,and observe pathological changes in the immune organs,20 mice were exposed in the ricin aerosol exposure system established in our lab previously;the mice were divided into 2 groups,control group and ricin aerosol exposure group.12 h,30 h and 48 h after exposure,mice were sacrificed,and then,trachea,lungs,thymus,spleen were collected.After they were sliced,the pathological changes were observed.The results showed that ricin aerosol would induce a series of pathological reactions in trachea,interstitial pneumonia,meanwhile,degeneration and necrosis of the thymocyte and splenocytes.Those results indicated that ricin aerosol can induce injury in immune barriers and provide some basic evidence for understanding pathological mechanism of injury caused by ricin aerosol poisoning.
