致病性坏死梭杆菌天然白细胞毒素毒性研究

摘　　要：为研究坏死梭杆菌白细胞毒素毒性及其致病机理,通过超滤截留技术从细菌培养物上清液分离获得坏死梭杆菌天然白细胞毒素,并通过细胞毒性试验、动物试验以及透射电镜观察等手段对分离获得的白细胞毒素的毒性进行研究。结果表明:坏死梭杆菌白细胞毒素对牛外周血白细胞具有细胞毒性作用,毒素毒性强弱与剂量呈正相关性,高浓度白细胞毒素能裂解白细胞,低浓度白细胞毒素则可诱导细胞凋亡。坏死梭杆菌白细胞毒素一方面能诱导宿主形成坏死病灶,另一方面也能诱导宿主产生对抗坏死梭杆菌感染的免疫保护力。In order to study the toxicity and pathogenic mechanism of leukotoxin of high pathogenic Fusobacterium necrophorum）,cytotoxicity test,microscopic observation and transmission electron microscope（TEM） were used to evaluate the toxicity of leukotoxin isolated from the culture supernatant of F.necrophorum.The results indicated that natural leukotoxin of F.necrophorum exhibits a high cytotoxicity to the bovine peripheral leukocytes in a dose dependent manner.The toxin induces apotosis at low concentration,and lyses the cell at higher concentration.Moreover,the natural leukotoxin can also induce necrosis lesions at the injecting site and protect the animal against experimental challenge with F.necrophorum.This study has laid theoretical foundation for understanding pathogenic mechanism of leukotoxin of F.necrophorum.
