摘 要:目的研究黄芪甲苷的抗脓毒症作用。方法采用盲肠结扎穿孔术(CLP)复制小鼠脓毒症模型,黄芪甲苷分别于小鼠CLP术前、术后1 h两次灌胃给药。结果黄芪甲苷显著提高实验性脓毒症小鼠的存活率,降低肺湿、干重比,降低血清MPO、NO、LDH、AST、ALT水平,降低肝脏组织中iNOS和IL-1β mRNA表达。黄芪甲苷还能提高肝脏组织中蛋白C(PC)mRNA和内皮细胞蛋白C受体(EPCR)mRNA表达,恢复受损的PC系统。结论黄芪甲苷对脓毒症小鼠具有保护作用,其机制与抗炎及上调PC系统有关。Objective To study the protective effect of astragaloside against sepsis in mice. Methods Cecum ligation perforation (CLP)operation was applied to establish sepsis model in mice . Preoperative and postoperative lavage administration of astragaloside was given to CLP mice. Results The survival rate of experimental septic mice was significantly increased after use of astragaloside, lung wet to dry weight ratio was also reduced, the level of serum MPO, NO, LDH, AST and ALT were lowered,and liver tissue of iNOS and IL-1 beta mRNA expressions were reduced. Astragaloside could improve PC mRNA and protein C receptor (EPCR)mRNA expressions in liver tissue and repair the damaged PC system. Conclusions Astragaloside possesses protective effect on septic mice and its mechanism is related to anti-inflammatory and the upregulation of PC system.
