摘 要:目的研究硫辛酸在神经细胞老化过程中对细胞内源性抗氧化分子谷胱甘肽(GSH)的作用。方法采用无血清培养神经母细胞瘤细胞建立实验性神经细胞老化模型。设立100μmol/L硫辛酸(LA)作用组和空白对照组,采用MTT法检测细胞存活率;比色法测定细胞内GSH含量。结果硫辛酸作用组5d、10d和15d时细胞内GSH含量分别为306.48μg/mgprot、240.72μg/mgprot、152.34μg/mgprot(同时期对照组分别为278.08μg/mgprot、181.94μg/mgprot、99.13μg/mgprot);随着细胞老化进程而下降,但LA实验组的细胞内GSH含量显著高于同时期对照组。结论硫辛酸能明显提升实验性神经细胞内GSH水平,增加细胞内源性抗氧化能力。Objective To study the effect of lipoic acid on glutathione(GSH)of neurons during experimental neuronal ag- ing. Methods The experimental model of neuronal aging was made by using the serum-free cultivated neuroblastoma cells. In this experiment, we established two groups: 100 μmol L-1 LA group and contrlo group, and then we detected the neuronal vitality(MTT method) and the GSH level(chromatometry). Results The GSH content in neurons was 306.48 t^g/mgprot, 240.72 t^g/mgprot and 152.34 t^g/mgprot in LA group, however, 278.08 t^g/mgprot, 181.94 t^g/mgprot and 99.13 b^g/mg- prot in control group on 5th, 10th and 15th, respectively. The GSH content in neurons in LA group was higher than that in control group with the decrease of neuronal aging. Conclusion Lipoic acid can significantly enhance the GSH level of the experi- mental nerve cells, which can increase endogenous antioxidant capacity of these cells.
