摘  要:为了解水稻品种多样性与稻瘟病菌群体变异的关系,采用rep-PCR分子指纹对分离自贵州省雷山县和湄潭县水稻间栽组合上的40个稻瘟病菌单孢菌株进行群体遗传结构分析.结果表明:40个单孢菌株分别扩增出5~9条谱带,大小为0.5~15 kb,80%左右的条带集中在1~10 kb;在0.74遗传相似水平下,供试菌株可划分为6个遗传宗谱,菌株间亲缘关系的远近与其寄主品种相关,3个间栽组合主栽品种上的11个菌株分属5个宗谱4个亚宗谱,净栽主栽品种上的14个菌株分属4个宗谱3个亚宗谱,而净栽糯稻上的8个菌株和间栽糯稻上的7个菌株则分属4个宗谱及3个宗谱3个亚宗谱.结论:间栽田块稻瘟病菌遗传宗谱组成较净栽田块丰富,水稻品种混合间栽有利于增加田间稻瘟病菌的遗传多样性,降低寄主遗传背景单一化对稻瘟病菌的定向选择风险.In order to know the relationship between rice variety diversity and rice blast population variation,forty isolates of M.grisea,which were collected from three mixed-planting combinations in Leishan and Meitan County in Guizhou Province,were analyzed for genetic structure based on rep-PCR fingerprinting.The result showed that all isolates were amplified 5~9 DNA bands,the size was 0.5~15 kb,but 80% concentrated in 1~10 kb.Six genetic lineages were determined at 74% genetic similarity,and the genetic relationship among isolates was related to their hosts.Eleven isolates from main varieties of three mixed-planting fields were classified into five genetic lineages and four sub-lineages.Fourteen isolates from monoculture fields were clustered into four genetic lineages and three sub-lineages.Eight isolates from glutinous rice of monoculture fields and seven isolates from mixture ones were merged into four lineages and three lineages and three sub-lineages,respectively.The results clearly indicated that the genetic structure composition in mixed-planting was richer than that in monoculture fields.The way to mix-planting rice cultivars is beneficial to the increasing of genetic diversity of M.grisea in fields and the reducing of directional selection risk on rice blast fungus due to simplification of host genetic background.
