甜瓜根分泌物介导自毒作用差异基因表达分析

摘　　要：以甜瓜品种"新银辉"为试验材料,利用甜瓜根系分泌物胁迫处理甜瓜幼苗,通过cDNA-AFLP技术分析甜瓜自毒作用相关基因及其表达情况。结果表明,利用63对引物组合获得约1200条TDFs(Transcription-derived fragments),平均每对引物组合扩增出15-30条TDF,长度在50-700bp之间。相关基因的表达模式可分为上调、下调、瞬时和持续表达4类。选取表达差异明显片段进行回收、测序、比对,获得42条同源性高的TDFs序列。甜瓜根系分泌物介导的自毒作用相关差异表达基因涉及能量代谢、物质合成与运输、逆境诱导和基因调控及信号传导等过程,分子调控机制较为复杂。In this research, allelopathic autotoxicity genes of ＂New Silverlit＂ melons treated with melon root exudates at seedling stage were analyzed by the cDNA AFLP technique. The results demonstrated that about 1200 transcript derived fragments （TDFs） were obtained via cl-）NA-AFLP with 63 primer pairs, and each of them produced 15-30 TDFs with 50-700 bp in length. The related genes expression patterns of melon could be divided into four categories which were the upward, downward, instantaneous and sustained expression. Forty two TDFs with high sequence homology were obtained by selecting, recovering and sequencing from the obviously different fragments. The functions of homologous genes included energy metabolism, material synthesis and transportation, stress inducing, gene regulation and signal transduction. The molecular regulation mechanism was complicated.
