摘 要:对云南昆明、楚雄、保山、德宏、临沧、玉溪、红河等地番木瓜、南瓜和罗汉果上获得的番木瓜环斑病毒(Papayaringspotvirus,PRSV)不同分离物进行了部分ci基因的克隆和序列分析,获得的部分ci基因长666nt,编码221个氨基酸。关于13个PRSV云南分离物部分ci基因核苷酸序列的同源性为80%。98%,氨基酸同源性为92%-100%。选取本文具有代表性的9个PRSV分离物与国内外12个PRSV分离物相比,核苷酸序列的同源性为79%-93%,氨基酸序列同源性为92%-99%。运用MEGA软件绘制的基于部分ci基因核苷酸序列的系统进化树表明,这些PRSV病毒可以分为两个大组,本文获得的9个云南分离物中的8个分离物与印度分离物(India—W—EU475877)聚为第1组,第Ⅱ组成员比较复杂,包括大部分的亚洲分离物及全部美洲分离物。Partial ci gene of Papaya ringspot virus (PRSV) isolates of Kunming, Chuxiong, Baos- ban, Dehong, Lincang, Yuxi and Honghe in Yunnan detected from papaya, pumpkin and Sirahia grosvenorii was cloned and sequenced. The partial ci gene obtained in this paper consists of 666 nt, encoding 221 amino acid. The nucleotide sequence of ci gene of 13 PRSV Yunnan isolates shared 80% to 100% identity with each other, the amino acid sequence showed 92% to 100% homologies. The nucleotide sequence of selected and representative 9 PRSV Yunnan isolates in this paper shared 79% to 93% identity with that of other isolates in worldwide and shared 92% to 99% amino sequence ho- mologies. Phylogenetic tree based on the nucleotide sequence of partial ci gene was constructed usingMAGE, and the result indicated that the tested PRSV isolates could be divided into two groups, group I includes 8 of 9 PRSV Yunnan isolates tested in this paper and one India isolate (India-W- EU475877), and members in group I1 were more complex, including and most of the Asia isolates and all of the America isolates.
