摘  要:目的探讨瓜蒌-薤白药对单用与配伍用于大鼠抗急性心肌缺血心肌损伤中对MAPKs信号通路的影响。方法将40只大鼠随机分为假手术(Sham)组、模型(I/R)组、瓜蒌(TK)组、薤白(AM)组、瓜蒌薤白(TKAM)组,通过结扎冠状动脉左前降支造成大鼠急性心肌缺血损伤模型,分别用药物进行干预,各组动物于不同试验时限(缺血30 min)时取出心脏。采用Western blotting法观察丝裂原活化蛋白激酶(mitogen activated protein kinases,MAPKs)信号通路细胞外信号调节激酶(extracellular signal-regulated kinase,ERK)、C-Jun氨基端激酶(C-Jun N-terminal kinase,JNK)、p38MAPKs磷酸化水平的影响,验证MAPKs信号在急性心肌缺血诱导心肌损伤发生发展中的作用;对心肌组织进行HE染色观察心肌坏死组织病理变化。结果对于大鼠抗急性心肌缺血模型,经电镜检查结果显示,配伍后的瓜蒌-薤白药对(1.875 g/mL)对大鼠急性心肌缺血所造成的心肌组织损伤具有相对显著的保护作用;经Western blot方法检验结果显示,与假手术组相比,在各组中以TKAM组P-p38及P-JNK的表达作用最弱,表明瓜蒌-薤白药对能有效抑制P-JNK及P-p38通路的蛋白进行表达;而通路P-ERK的表达结果却与其他两条通路相反,由此可以推论ERK信号通路的激活可能对心肌损伤具有保护作用。结论瓜蒌薤白配伍后抗心肌组织损伤的效果更显著;3种丝裂原活化蛋白激酶(MAPKs)在瓜蒌薤白药对抗急性心肌缺血过程中发挥着不同的作用。Objective To explore the influence of single use and compatibility use of Trichosanthes kirilowii Maxim and Allium macrostemon Bge. on the mitogen activated protein kinases (MARKs) signal pathway against rat acute myocardial ischemic myocardial damage. Methods 40 rats were randomLy divided into the sham(Sham) group,model (I/R) group, Trichosanthes kirilowii Maxim (TK) group, Allium macrostemon Bge. (AM) group and Trichosanthes kirilowii Maxim and Allium macrostemon Bge. (TKAM) group. The lgation of the left anterior descending coronary artery was performed to establish the rat damage model of acute myocardial ischemia. The vari- ous groups were respectively given the drug intervention. The rat heart was taken at different experiment time limit (ischemia for 30 min). The Western blot method was adopted to observe the influences of extracellular signal regulated kinase (ERK),C-Jun amino end kinase (JNK),p38MAPKs phosphorylation level in the mitogen activated protein kinase (MAPKs) signal pathway for verifying the role of MAPKs signal in the occurrence and development of acute myocardial ischemia induced myocardial damage;in addition,the my- ocardial tissues were performed the HE staining for observing the histopathological changes of myocardial necrosis tissues. Results The scanning electron microscopy (SEM) results showed that the compatibility (1. 875 g/mL) of Trichosanthes kirilowii Maxim and Allium macrostemon Bge. had relative signficant protective effect on the rat myocardial tissue damage caused by acute myocardial ischemia;the Western blot results showed that compared with the Sham group, among other groups,the P-p38,P-JNK expression effect in the TKAM group was weakest, indicating that Trichosanthes kirilowii Maxim and Allium macrostemon Bge. could effectively inhibit P - JNK,P-p38 channel protein expression;but the expression results of pathway P- ERK was contrary to other two pathways,it could be inferred that the activation of the ERK signaling pathways mi
