摘  要:目的探讨苯并芘(benzo-a-pyrene,BaP)导致胎盘绒毛组织细胞凋亡的作用机制及维生素E的保护作用。方法选择性成熟ICR小鼠150只(雌鼠100只,雄鼠50只),随机分为A、B、C、D、E共5组,每组各30只,雌雄分开饲养。A组予BaP 2 mg/(kg·d);B、C、D组予BaP同时,分别予维生素E 10、20、40 mg/(kg·d);E组灌胃橄榄油。15 d后雌雄鼠同笼,第2 d发现精液栓者为受孕,将受孕鼠继续灌胃7 d后处死,取胎盘组织测定Bax、Bcl-2基因表达。结果 BaP能使Bax基因蛋白及mRNA的表达量升高,Bcl-2基因蛋白及mRNA的表达量下降,Bax/Bcl-2的比值升高,A组与E组比较差异均有统计学意义(P<0.05);与A组比较,予维生素E的各组Bax基因表达量降低而Bcl-2表达量升高,BaP/Bcl-2比值降低,差异均有统计学意义(P<0.05)。Bax、Bcl-2基因蛋白及mRNA表达量存在一定相关性(r=-0.951,P=0.002;r=-0.825,P=0.021)。结论 BaP通过升高Bax与降低Bcl-2基因表达量使胎盘绒毛组织细胞凋亡,而维生素E可阻止这一途径而抑制其凋亡。Objective To explore the mechanism of action of placental villus apoptosis induced by Benzopyrene (BaP) and the effect of Vitamin E.Methods A total of 150 viripotent ICR mice (100 males,50 females) were randomly divided into five groups (n =30),and female mice and male mice were raised respectively.Group A was lavaged with BaP 2 mg/(kg · d); group B,C and D were lavaged with BaP 2 mg/(kg · d) and Vitamin E 10,20 and 40 mg/(kg · d) at the same time; group E was layaged with olive oil.After lavage for 15 d males were copulated with females,and gestational age was determined by the presence of a vaginal semen plug at the second day.The pregnant mice were killed after continued lavaging for 7 d.The gene expressions of Bax and Bcl-2 in placental tissue were detected.Results BaP could induce the increase of Bax expression,reduction of Bcl-2 expression,and increase of the Bax/Bcl-2 ratio,and the differences between group A and group E were statistically significant (P <0.05); compared with group A,the Bax expressions were reduced,Bcl-2 expressions were decreased,and BaP/Bcl-2 ratios were increased in the groups lavaged with Vitamin E,and the differences were also statistically significant (P < 0.05).The genes of Bax and Bcl-2 in the protein and mRNA had a certain correlation (r =-0.951,P =0.002 ; r =-0.825,P =0.021).Conclusion BaP can induce placental villus apoptosis by increasing the Bax expression and decreasing the Bcl-2 expression,while Vitamin E can stop the progress and suppress apoptosis.
