增殖性糖尿病视网膜病变玻璃体中转化生长因子β_2的定量分析

摘　　要：目的 :通过对增殖性糖尿病视网膜病变 (proliferative diabetic retinopathy,PDR)患者玻璃体中转化生长因子 β2(TGF-β2 )含量的研究 ,探讨 TGF-β2 在 PDR病理过程中的作用。 方法 :应用酶联免疫吸附测定法对 10例 (10只眼 )正常人及2 4例 (2 4只眼 ) PDR患者玻璃体中 TGF- β2 的含量进行检测和分析。 结果 :10例正常人玻璃体中 ,TGF- β2 的含量为 (1.2 7±0 .6 9) ng/ml;2 4例 PDR患者玻璃体中 ,TGF- β2 的含量为 (4 .89± 3.5 1) ng/ml。两组比较 ,PDR组的玻璃体中 TGF- β2 的含量明显升高 ,差异非常显著 (P<0 .0 1)。将糖尿病视网膜病 期及 期 14例 (14只眼 )患者列为一组 ,10例 (10只眼 ) 期患者另列一组 ,玻璃体中 TGF- β2 的含量分别为 (6 .0 7± 4.0 5 )及 (2 .84± 1.72 ) ng/m l,两组间比较有显著差异 (P<0 .0 5 )。它们分别与正常对照组比较也有非常显著差异 (P<0 .0 1)和显著差异 (P<0 .0 5 )。结论 :PDR患者玻璃体中 TGF- β2 的含量升高 ,提示 TGF- β2 在Objective: To compare the levels of transforming growth factor β 2(TGF β 2) in the vitreum of patients with proliferative diabetic retinopathy (PDR)and normal subjects and to evaluate the role of TGF β 2 in the pathological course of PDR. Methods: Using ELISA, the total TGF β 2 levels in the undiluted 24 vitreous samples of patients with PDR and 10 normal subjects were analysed quantitatively. Results: There was a very significant difference in total TGF β 2 between the normal group [ (1.27± 0.69) ng/ml, n =10] and the PDR group [(4.89±3.51) ng/ml , n =24] ( P <0.01).The patients of 24 PDR were divided into 2 groups according to clinical findings:14 patients of DR stage Ⅳ and Ⅴ as one group, and 10 patients of DR stage Ⅵ as the other group. The total levels of TGF β 2 in the vitreum were (6.07±4.05) ng/ml and (2.84±1.72) ng/ml, respectively ( P <0.05). There was a significant difference also between above two groups and normal subjects ( P <0.01 and P <0.05, respectively). Conclusion: The results indicate that increased level of total TGF β 2 in the vitreum of patients with PDR may play an important role in the pathological course of PDR. [
