摘 要:目的:研究体外实验中阳和化岩汤为代表的温阳散结中药及拆方对乳腺癌癌前病变的MCF-10AT细胞的生长抑制及诱导凋亡作用。方法:采用乳腺癌癌前病变的MCF-10AT细胞,分组阳和化岩汤组及拆方温阳组、活血组、化痰组、解毒组、三苯氧胺组、空白对照组7组,MTT法观察24h、48h、72h药物对细胞的生长抑制率,流式细胞仪测定药物干预72h后细胞凋亡率和细胞周期的变化。结果:①TAM组、阳和化岩汤组及拆方各组对MCF-10AT细胞具有明显生长抑制作用,呈现时间依赖效应(P<0.05)。②药物干预72h各组细胞凋亡率明显增高,阳和化岩汤组与三苯氧胺组诱导凋亡率对比无显著性差异(P>0.05);但拆方各组细胞凋亡率低于三苯氧胺组(P<0.05)。③与空白对照组相比,三苯氧胺、阳和化岩汤组及拆方解毒组处理可导致MCF-10AT细胞的S期阻滞(P<0.05);阳和化岩汤组、三苯氧胺细胞G2/M期比例下降(P<0.05);G0/G1期细胞呈相对平稳趋势,各组无显著差异。结论:阳和化岩汤组与三苯氧胺组诱导乳腺癌癌前病变MCF-10AT细胞凋亡能力相近,拆方各组低于三苯氧胺组。温阳散结法对乳腺癌癌前病变细胞株有效生长抑制,呈现时间依赖效应;诱导细胞凋亡,阻滞细胞周期于S期。从增殖抑制、诱导凋亡的角度验证温阳散结中药在乳腺癌癌前病变的阻断治疗中的作用机制,适用于乳腺癌癌前病变的预防治疗。Objective: To research the inhibitive effect of Warm-yang-disperse-plegm therapy delegated by Yanghe Decoction Huayan on separate growth inhibition and induction of apoptosis in precancerous lesions of breast cancer MCF-10AT cells.Method: We divided the subjects into Yanghe Huayan Decoction group and Warm-yang group,blood-activating and stasis-resolving group,resolving-phlegm group,tamoxifen group and blank control group.MTT method was used to observe the 24h,48h,72h of medicine on cell growth inhibition rate,measured by flow cytometry 72h after medicine intervention rate of apoptosis and cell cycle changes.Results: TAM group,Yanghe Huayan Decoction group and its separate group of MCF-10AT cells with obvious growth inhibition,showing time-dependent effect(P &lt;0.05).Medicine intervention 72h apoptosis rate was significantly higher,There was no significant difference between Yang group of rock soup and tamoxifen group induced apoptosis rate(P &gt; 0.05);demolition party groups apoptosis rate was less than tamoxifen group(P &lt; 0.05).Compared with the blank control group,tamoxifen,YangheHuayan Decoction group process could lead to the MCF-10AT cells in S stage(P &lt; 0.05);YangheHuayan Decoction group,tamoxifen cells in G2/M stage decreased(P &lt; 0.05);G0/G1 phase cells was relatively stable trends,and each group was no significant difference.Conclusion: YangheHuayan Decoction group and the tamoxifen group have the similar ability to induce precancerous lesions apoptosis breast cancer MCF-10AT,of This therapy is suituble for the preventive treatment of such disease.
