摘  要:目的:探明树舌多糖GF(Ganoderma appanatum polysaccharides GF,GAPSGF)对SMMC7721肝癌细胞增殖及CyclinE基因mRNA表达的影响。方法:用80、40、20、10、5、2.5、1.25、0.625μg/mL树舌多糖GF分别处理肝癌细胞SMMC7721,48h后MTT法检测树舌多糖GF对SMMC7721细胞增殖的抑制情况;实时定量聚合酶链反应(Real-Time PCR)检测CyclinE mRNA表达。结果:树舌多糖GF对SMMC7721细胞具有增殖抑制作用。Real-time PCR结果显示,CyclinE mRNA的表达量明显低于模型组。结论:树舌多糖GF可下调CyclinE mRNA的表达,从而抑制SMMC7721肝癌细胞的增殖。Objective: To study the effects of Ganoderma appanatum polysaccharides GF(GAPSGF) on proliferative inhibition and CyclinE mRNA expression in hepatoma cell line SMMC7721.Methods: The effect of GAPSGF on SMMC7721 cells proliferation was studied by using MTT.Real-time PCR was used to examine the expression of mRNA of CyclinE.Results: GAPSGF could effectively inhibit proliferation of SMMC7721 cells.GAPSGF group could significantly reduce the expressions of mRNA of CyclinE in SMMC7721 cells as compared with those in control group.Conclusion: GAPSGF can inhibit cells proliferation through down-regulating CyclinE expression in SMMC7721 cells.
