针刺对实验性脑出血大鼠脑组织GDNF和nestin表达的影响

摘　　要：目的:探讨急性脑出血后胶质源性神经生长因子(GDNF)和巢蛋白(nestin)的动态表达及针刺的干预作用。方法:选取健康的雄性Wistar大鼠160只,随机分为模型组、模型加针刺组(针刺组)、模型加忆立福组(西药组)3组。每组又分为6h、24h、48h、72h和168h5个时相点,每个时相点有10只大鼠,空白对照组10只,其余制备脑出血大鼠模型,运用免疫组化等方法检测各组大鼠脑组织中GDNF和nestin阳性细胞的表达以及针刺治疗对它们的影响。结果:模型组6h出现GDNF阳性细胞表达增加,1天时达高峰,7天时仍有表达,针刺组与模型组、西药组相比有统计学意义(P<0.01);模型组6h出现nestin阳性表达增加,2天时达高峰,7天时仍有表达,针刺组与模型组、西药组相比较,差异有统计学意义(P<0.01)。结论:针刺通过调控神经营养因子GDNF和神经干细胞nestin的表达,增加神经干细胞增殖分化能力,这可能是针刺"百会"透"曲鬓"改善脑出血大鼠神经功能缺损症状和大鼠脑组织超微结构,治疗急性脑出血神经重塑的机制之一。Objective： To study the dynamic expression of GDNF and nestin after acute intercerebral hemorrhage（ICH） in rats and the influence of acupuncture on it.Methods： 160 healthy Wistar male rats were selected.The rats were divided into three groups randomly： model group,pinprick group,western medicine group,30 rats in each group.Each group was divided into five time phase point of 6 hours,24 hours,48 hours,72 hours,168 hours again,10 rats in each group.Experimental ICH models were made by injecting antologous blood using stereotactic method.Another 10 rats were assigned as the matched blank group.The expression of GDNF and nestin positive cells in intracerebral hemorrhage rats of each group were measured by immunohistochemistry methods respectively and the influence of the expression by acupuncture was observed.Results： 6h GDNF positive cells the model group appeared to increase;got peak at 1d,and kept expression till 7d.Pinprick group and model group,compared with western medicine group differences were statistically significant（P ＆lt; 0.01）.In model group,nestin expression appeared to increase at 6h,reached its peak at 2d,kept expression till 7d.Compared with madel group and western medicine group,the differences were statistically significant in acupuncture group（P ＆lt; 0.01）.Conclusion： By regulating neurotrophic factor GDNF and nestin’s expression,acupuncture treatment can improve the ability of neural stem cells to the proliferation and differentiation.This may be the treatment of acupuncture＂Baihui＂through＂Qubin＂made improvement of neurologic impairment symptoms of intracerebral hemorrhage and ultrastructure stracture of rats brain tissue,and it is one of these mechanisms for acute neural remodeling in intracerebral hemorrhage.
