HMG联合GnRH拮抗剂对子宫内膜细胞增殖及整合素表达的影响

摘　　要：目的:尿促性素单独或联合GnRH拮抗剂刺激对子宫内膜上皮细胞增殖及整合素mRNA表达的影响。方法:用不同浓度的尿促性素单独或联合GnRH拮抗剂处理Ishikawa细胞,通过MTT法检测细胞增殖变化,用realtime PCR法检测整合素mRNA表达变化。结果:(1)尿促性素浓度低于100IU/ML时,对Ishikawa细胞的增殖没有明显影响(P>0.05),而尿促性素浓度浓度为1000IU/ML时,Ishikawa细胞增殖能力显著降低(P<0.05);(2)尿促性素浓度低于100IU/ML时,对Ishikawa细胞整合素β3 RNA表达没有明显影响(P>0.05),而尿促性素浓度浓度为1000IU/ML时与对照组比较,Ishikawa细胞整合素β3RNA表达明显受到抑制(P<0.05);(3)添加GnRH拮抗剂处理细胞后,没有改变上述实验结果。结论:低剂量的尿促性素联合GnRH拮抗剂刺激对子宫内膜上皮细胞增殖及整合素mRNA表达无影响,微刺激联合拮抗剂方案促排卵可能不改变子宫内膜容受性。Objective： To study the effect of HMG alone or with GnRH antagonist on the cell proliferation and integrinβ3 mRNA expression of human endometrial cancer cell Ishikawa.Methods： The Ishikawa cells were stimulated by different concentration of HMG alone or combine with GnRH antagonist,and determining cell proliferation by MTT assay,detecting integrinβ3 mRNA expression through real time PCR method.Results：（1） when Menotrophin concentration below 100IU/ml,there was no obvious impact on the Ishikawa cell proliferation capacity（P0.05）,and it was significant reduced when the concentration up to 1000 IU/ml（P0.05）;（2）when HMG concentration below 100IU/ml,there was no obvious effect on integrinβ3 mRNA expression of Ishikawa cells（P0.05）,but it was significant inhibited when the concentration up to 1000IU/ml compared with control groups（P0.05）;（3） the above results didn’t change after adding in GnRH antagonist.Conclusion： Low concentration of HMG combined with GnRH antagonist could not alter the cell proliferation capacity and integrinβ3 mRNA expression of Ishikawa cells,mild stimulate ovulation protocol with GnRH antagonist may not affect endometrial receptivity.
