摘 要:黄连木(Pistacia chinensis Bunge)是一种重要的多年生木本油料植物,建立其高效的基因转化体系是进行基因工程育种的重要环节。在以抗生素为筛选标记的基因转化中,确定适宜的筛选压力是提高基因转化效率的前提。本研究以黄连木子叶为材料,探讨了四种抗生素对其分化诱导及生长的影响。结果表明,四种抗生素对黄连木子叶分化诱导及生长的影响具有明显的差异。随着抗生素浓度的不断提高,子叶分化诱导频率呈逐渐下降的趋势,当卡那霉素浓度达到50 mg/L、潮霉素浓度达到10 mg/L时,子叶分化的诱导基本上被抑制,表现出较好的效果;与其相比,头孢霉素、氨苄青霉素二种抗生素对芽的诱导抑制作用相对较弱,这说明卡那霉素、潮霉素可以作为一种有效的筛选转基因黄连木的抗生素筛选标记。研究结果为下一步进行黄连木的遗传转化研究奠定了基础。Pistacia chinensis Bunge is a perennial woody energy plant, for which establishing an efficient genetic transformation system plays an important role in gene engineering breeding. The optimal screening pressure by antibiotics could lead to the high genetic transformation efficiency. The effects of four antibiotics on the growth and differentiation of cotyledons were studied. Four kinds of antibiotic researches showed the obvious differences between growth and differentiation of cotyledons. The differentiation of cotyledon was decreasing when increasing the concentration of antibiotics. The optimum concentrations of kanamycin and hygromycin are 50 mg/L and 10 mg/L respectively, which had a significantly inhibition on the cotyledon differentiation. However, it would not affect the growth of transformed plantlets. When compared with kanamycin and hygromycin, cephamycin and ampicillin presented a lower inhibitory effect on the induction of shoots. All the results above indicated that kanamycin and hygromycin are the effective antibiotic selection markers in transgenic pistacia chinensis Bunge screening, which could pave a way for the further study on genetic transformation ofpistacia chinensis Bunge.
