摘 要:目的:观察miR-143在具有肥胖及糖尿病表型的ob/ob小鼠与正常对照组C57/BL6J小鼠肠系膜脂肪、肝脏组织及骨骼肌中的表达情况。方法:选择16周龄的ob/ob小鼠与C57/BL6J小鼠各12只,采用实时荧光定量聚合酶链反应技术(qRT-PCR)检测两组小鼠肠系膜脂肪、肝脏组织及骨骼肌的表达水平。结果:miR-143在ob/ob小鼠肠系膜脂肪、肝脏组织中表达上调(P<0.05),而在骨骼肌中无显著性差异(P>0.05);ob/ob小鼠肠系膜脂肪miR-143的表达与HOMA-IR呈正相关。结论:miR-143可能参与了ob/ob小鼠肥胖及胰岛素抵抗(IR)的发生。Objective: To investigate the expression and significance of miR - 143 in mesenteric fat, liver and skeletal muscle in oh/oh mice. Methods: Twelve ob/ob mice and 12 C57/BL6J mice of 16 weeks old were select- ed respectively and qRT - PCR was used to detect the expression level of miR - 143 in the mesenteric fat, liver and skeletal muscle tissue. Results: The expression of miR - 143 in the mesenteric fat and liver was up - regulated in oh/oh mice ( P 〈 0.05 ), but no significant difference was found in skeletal muscle between the two groups ( P 〉 0.05 ). miR- 143 expression levels in mesenteric fat were positively correlated with HOMA -IR. Conclusion: miR - 143 may be involved in the occurrence of obesity and insulin resistance in oh/oh mice.
