摘 要:目的研究浒苔(绿藻门,石莼科,浒苔属)正丁醇萃取相的化学成分及其对CCl4和D-GalN诱导的小鼠肝损伤的保护作用。方法采用硅胶柱色谱、Sephadex LH-20柱色谱、高效液相色谱、薄层制备等方法进行分离纯化;利用MS、1 H NMR、13 C NMR波谱分析结合文献对照鉴定化合物结构;用CCl4和D-GalN诱导小鼠肝损伤造模,检测小鼠肝脏指数,血清中AST、ALT水平,肝脏中总SOD、MDA水平来探讨其对肝损伤的保护作用。结果从浒苔正丁醇萃取相分离鉴定出了6种化合物:对羟基苯甲酸甲酯,2-脱氧尿嘧啶核苷,尿苷,尿嘧啶,次黄嘌呤,腺嘌呤;活性实验方面,与模型组相比,中剂量组、高剂量组均可以显著抑制肝脏指数,血清AST、ALT以及肝脏中MDA的水平,各剂量组对受损肝脏SOD水平都有明显的恢复作用。结论浒苔正丁醇相化学成分以核苷类化合物为主,而且可以明显减轻由CCl4和D-GalN诱导的小鼠急性肝损伤。Objective To investigate the chemical composition and protective effect of the n-butanol ex- tracts from Enteromorpha prolifera on CC14 and D-GalN induced liver injury in mice. Methods The compounds were isolated and purified by silica column chromatography, Sephadex LH-20 column chro- matography, high performance liquid chromatography and thin layer chromatography for preparation. Their structures were identified with the spectroscopic analysis of MS, 1H NMR,13 C NMR and compari- son with the data from literatures; The model was established on CC14 and D-GalN induced liver injury in mice. The liver index, AST and ALT levels in serum, SOD and MDA levels in liver were detected to investigate the protective effect of n-butanol extracts from Enterornorpha prolifera on CC14 and D-GaIN induced liver injury in mice. Results Six compounds were isolated from the n-butanol extracts of Entero- rnorpha prolifera: methylparaben, 2-deoxyuridine, uridine, uracil, hypoxanthine, and adenine; In the activity experiments, compared with model group, both the moderate dose and high dose groups could significantly inhibit the exaltation of the liver index, AST and ALT levels in serum, as well as the MDA level in liver. All of the groups could significantly restore the SOD activity in the injured liver. Conclusion Nucleosides were the main components of the n-butanol extracts from Enteromorpha prolif- era, which can significantly alleviate acute liver injury induced by CC14and D-GalN.
