摘 要:目的:考察灯台叶总生物碱提取物中鸭脚树叶碱和瓦莱萨明碱在大鼠肠道的吸收情况,为其合理用药提供生物药剂学依据。方法:采用大鼠在体肠灌流模型,应用RP-HPLC法测定鸭脚树叶碱及瓦莱萨明碱的含量,研究不同浓度总生物碱中鸭脚树叶碱及瓦莱萨明碱在大鼠不同肠段的吸收情况。结果:相同浓度下鸭脚树叶碱及瓦莱萨明碱在各肠段的有效渗透系数(Peff*)均>1.0,依次为十二指肠>结肠>空肠>回肠,各肠段之间有显著性差异(P<0.05);不同浓度同一肠段,鸭脚树叶碱及瓦莱萨明碱在各肠段的Peff*值无显著性差异。结论:鸭脚树叶碱及瓦莱萨明碱在大鼠肠道四个肠段内均有吸收,尤其在十二指肠、结肠段吸收较好;药物浓度对鸭脚树叶碱及瓦莱萨明碱的Peff*值无显著性影响,初步判断其吸收机制为被动扩散。OB.IECIVE To investigate the rat intestinal absorption behavior of pierinine and vallesamine in total alkaloids of leaves of Alstonia scholaris, and provide biopharmaceutic basis for reasonable application. METHODS The intestinal perfusion method was applied to the intestinal absorption of picrinine and vallesamine in different total alkaloid concentrations. The con- tent of picrinine and vallesamine in the perfusate sample was determined by RP-HPLC. RESULTS At the same concentra- tions, both the effective permeability coefficients (Po) of picrinine and vallesamine were higher than 1.0 and had significant difference (P〈0. 05) in four different segments, which showed highest in the duodenum, followed by colon, and then jejunum and ileum; while there was no significant difference of different concentrations from Pef. of pierinine and vallesamne in the same intestinal segments. CONCLUSION Picrinine and vallesamine can be well absorbed in intestinal tract of rat with better absorption in duodenum and colon. The absorption mechanism of picrinine and vallesamine might be passive diffusion as drug concentration has no significant impact on Po of picrinine and vallesamine.
