摘 要:以滨梅Prunus maritima Marshall茎段为外植体,研究了培养基和培养条件对滨梅组培苗玻璃化现象的影响,探索滨梅组培苗的最佳培养方法。结果表明,导致滨梅组培苗玻璃化的主要因素是培养基中的TDZ、蔗糖和琼脂浓度以及培养温度和光照时间,其中由正交试验结果显示,影响滨梅组培苗玻璃化的因子由大到小依次是TDZ浓度〉培养温度〉蔗糖浓度〉光照时间〉琼脂浓度,综合各试验结果,最后筛选出克服滨梅组培苗玻璃化的最优培养条件为TDZ浓度为0.5mg/L,蔗糖浓度为3%,琼脂浓度为0.6%,培养温度为25℃,光照时间为12~16小时/天(3000lx),在此条件下滨梅组培苗的繁殖系数较高,玻璃化率较低,植株生长健壮。Stem segments of beach plum ( Prunus maritima Marshall) were cultured in MS medium to observe the influence of culture medium and condition on vitrification of plantlets. The results showed that the main factors leading to vitrification of beach plum plantlets were concentrations of TDZ, sucrose and agar in the culture medium, also culture temperature and photoperiod. The orthogonal experiment results showed that the impact factors on vitrification of plantlets in descending order is TDZ concentration〉 culture temperature〉 sucrose concentration〉 illumination time〉 agar concentration. Based on single factor and orthogonal tests, we concluded that the optimized culture condition was at TDZ concentration of 0. 5 mg/L, sucrose concentration of 3 %, agar concentration of 0. 6M, temperature at 25 ℃, and irradiation time of 12-16 h/d (3 000 lx). This condition delivered higher proliferation coefficient of plantlets, the lower vitrification rate and the stronger growth of plant.
