摘 要:本试验旨在探讨纳米蒙脱石(MNC)驱铅对Caco-2细胞增殖率、LDH活性及细胞形态的影响。结果表明:随铅浓度的提高,其抑制细胞增殖率也越强,当铅浓度达40、80 mg/L时,铅抑制Caco-2细胞的增殖率显著(P<0.05);随时间延长,铅抑制细胞增殖率的作用越明显,并呈一定的时效关系;添加0.5%MNC能有效逆转因铅导致的低细胞增殖率,且其30 min解铅毒功效强于60 min;30 min反应中,大于40 mg/L铅浓度引起Caco-2细胞培养液中的LDH活性显著高于对照组(P<0.05);120 min反应中,大于20 mg/L铅浓度引起的细胞LDH活性显著高于对照组(P<0.05)。但0.1%和0.5%MNC分别添加到含40 mg/L铅的细胞培养液中,30 min反应后细胞LDH活性都显著低于对照组(P<0.05),且0.5%MNC组显著低于0.1%MNC组(P<0.05)。此外,40 mg/L铅攻毒30 min后,发现Caco-2细胞出现萎缩,呈不规则圆形,细胞间质明显,但同时添加40 mg/L铅+0.5%MNC后观察,Caco-2细胞维持正常形态。结果提示,MNC能有效吸附培养液中的铅,阻断铅进入Caco-2细胞,对动物细胞不会产生任何人毒副作用。This study was conducted to evaluate the effects of montmorillonite nanocomposite (MNC) as a Pb-sorbent on caco-2 cell proliferation rate, LDH activity and cellular morphology in vitro. The results showed that different concentration PBNK have no difference on cell proliferation rate and releasing LDH in different time (P〈0.05). 40, 80 mg/LPb2+ significantly increased releasing LDH (P〈0.05) morphology in time-dose dependent manner. Toxicity , reduced cell proliferation rate (P〈0.05) and damaged the cellular reaction to caco-2 cells of 20, 40, 80 mg/L Pb2+ in 60 minutes were significantly higher than those of 30 minutes (P〈0.05). The addition of 0.5% MNC in cell cultures could eliminate the low cell proliferation rate resulted by Pb2+, and detoxification effect of MNC in 30 minutes were better than in 60 minutes, but there was no obvious difference (P〉0.05). Above 40 mg/L Pb2+ in 30 minutes significantly increased LDH activities cell culture compared to control group (P〈0.05). While 0.1 and 0.5% PABB added into 40 mg/L Pb^2+ cell culture respectively, LDH activities in 30 minutes reduced obviously (P〈0.05), LDH activities of 0.5% MNC group decreased significantly compared with 0.1% MNC group (P〈0.05). In addition, it was showed that the Caco-2 cells appeared atrophic, irregular, and obvious mesenchymal cells in 40 mg/L Pb2+ 30 min attack poisoning, but Caco-2 cells maintain normal morphology after adding 0.5% MNC. These results indicated that MNC is a potential lead- sorbent through adsorbing the Pb2+ of cell culture and blocking Pb^2+ into Caco-2 cells.
