加味黄连阿胶汤对顺铂肾毒性大鼠肾组织Ⅲ型胶原表达的影响

摘　　要：目的:观察加味黄连阿胶汤对顺铂致肾损伤大鼠Ⅲ型胶原(collagenⅢ,ColⅢ)表达的影响。方法:将SPF级SD雄性大鼠60只随机分为正常对照组、模型组、硫代硫酸钠治疗组、加味黄连阿胶汤低剂量组、高剂量组,每组12只。正常对照组大鼠给予0.9%氯化钠溶液尾静脉注射,其余4组大鼠给予顺铂(DDP)尾静脉注射,5 mg·kg-1,1次/周,连续3周。硫代硫酸钠治疗组同时给予10%硫代硫酸钠注射,600 mg·kg-1;加味黄连阿胶汤低、高剂量组第1次尾静脉注射DDP后根据体质量分别给予0.5、1.5 g·mL-1加味黄连阿胶汤灌胃;正常对照组和模型组给予0.9%氯化钠溶液灌胃,1次/d。连续给药8周后剖取大鼠肾脏,固定、切片后,行免疫组织化学染色。结果:与模型组比较,加味黄连阿胶汤低、高剂量组及硫代硫酸钠治疗组ColⅢ的表达明显减弱,平均积分光密度值显著降低(P<0.05或P<0.01),且加味黄连阿胶汤低、高剂量组优于硫代硫酸钠治疗组(P<0.05),但加味黄连阿胶汤低、高剂量组组间差异无统计学意义(P>0.05)。结论:加味黄连阿胶汤可使顺铂肾毒性大鼠肾组织ColⅢ沉积明显减少,肾损害明显减轻。Objective： To investigate the effect of Supplemented Coptis and Ass Hide Glue Decoctionon on ColⅢ of cis- platin（DDP）- induced nephrotoxicity in rats. Methods： Maleness Sprague-Dawley rats were randomly divided into normal con- trol, model group, sodium thiosulfate group, and groups of Supplemented Coptis and Ass Hide Glue Decoctionon at low dose and large dose, 12 rats in each group. Cisplatin（5 mg.kg-1, 1 g.L-1） was injected through tail-vein at the same time each week for 3 weeks to all groups, but the model, which was given with normal sodium （1 mL. 100 g-1）. By the end of test, kindey tis- sues were taken out, wax-wrapped, immunohistochemistry stained to detect the expression of Col Ⅲ. Results： Compared with the model group, the expression of Col Ⅲ was weaker in sodium thiosulfate group, groups of Supplemented Coptis and Ass Hide Glue Decoctionon at low dose and large dose （P〈0.05 or P〈0.01）; The expression of Col Ⅲ in groups of Supplemented Coptis and Ass Hide Glue Decoctionon at low dose and large dose were weaker than that of model group （P〈0.05）; The dif- ference between the groups of Supplemented Coptis and Ass Hide Glue Dec, octionon at low dose and large dose was without significance （P〉0.05）. Conclusion： Supplemented Coptis and Ass Hide Glue Decoctionon can reduce the CoⅢ deposition in the kidney of rats with nephrotoxicity induced by cisplatin, and mitigate the ：renal damages significantly.
