摘 要:目的:探讨花色素苷(Anth)功能饮料对正常及环磷酰胺处理大鼠免疫功能的调节作用。方法:从黑米中提取Anth并将其调配成饮料,连续喂养大鼠6周后,测定脾脏和胸腺的重量及其系数;以四甲基偶氮唑盐(MTT)法检测由刀豆蛋白A、脂多糖诱导的脾T、B淋巴细胞的增殖活性;用生化方法测定腹腔巨噬细胞(PMΦ)、血清、肝、脾、肾、胸腺组织细胞中乳酸脱氢酶(LDH)、酸性磷酸酶(ACP)的活性;摄取中性红试验检测PMΦ的吞噬功能。结果:Anth饮料使正常大鼠脾湿重显著增加(P<0.01),并抑制环磷酰胺所致的脾萎缩(P<0.01);显著提高正常大鼠脾T、B淋巴细胞的增殖活性(均P<0.01),同时对环磷酰胺处理后大鼠脾T、B淋巴细胞降低的增殖活性具有明显改善作用(均P<0.05);显著提高正常大鼠PMΦ内LDH活性(P<0.01),而且能抵抗环磷酰胺所致的LDH活性(PMΦ、血清、肾、胸腺)和ACP活性(PMΦ、肝、脾、肾、胸腺)的降低(P<0.05或P<0.01);同时对环磷酰胺导致的大鼠PMΦ吞噬能力的降低具有明显上调作用(P<0.01)。结论:Anth饮料对大鼠的免疫功能具有增强作用,并能在一定程度上拮抗环磷酰胺的免疫抑制作用。Objective: To explore the regulation of anthocyanin (Anth) functional beverage on immunologi-cal function in normal and cyclophosphamide-treated rats. Methods: Anth was extracted from black rice fraction and was concocted an Anth beverage. After rats were administered with Anth beverage for 6 weeks, the wet weight of spleen and thymus were measured and their weight indexes were calculated, and the colorimetric MTT was used to analyze the proliferative activity of T or B lymphocyte in spleen which had been stimulated with ConA or LPS, the activities of lactate dehydrogenase (LDH) and acid phosphatase (ACP) in peritoneal macrophages(PMФ), serum, liver, spleen, kidney and thymus were determined by biochemical methods, and the capability of PMФ in phagocytosis of neutral red were measured by colorimetric method, too. Results: After Anth beverage was administered to rats, the results were observed as follows. The spleen weight increased remarkably (P〈0.01) in normal rats and spleen atrophy induced by cyclophosphamide were inhibited significantly(P〈0.01). The prolif- erative activity of T and B lymphocyte in spleen were increased significantly (P〈0.01 respectively) in normal rats, and the decrease of the proliferative activity of T and B lymphocyte in spleen of rats due to cyclophosphamide were notably antagonized (P〈0.05 respectively). The LDH activity in PMap of normal rats were enhanced significantly (P〈0.01), and the decrease of LDH activities in PMqb, serum, kidney and thymus as well as the reduction of ACP activities in PMФ, liver, spleen, kidney and thymus of cyclophosphamide-treated rats was repressed remarkably (P〈0.05 or P〈0.01 respectively). And the decrease of the capability of PMФ phagocytizing the neutral red due to cyclophosphamide was also antagonized significantly (P〈0.01). Conclusion: Anth beverage can enhance im- mune function and antagonize the immunosuppressive effects of cyclophosphamide to some extent in rats.
