摘 要:目的以实验大鼠为研究对象,寻找丙烯酰胺(acrylamide,ACR)神经损伤的生理生化基础。方法 48只Wistar大鼠随机分为对照组(腹腔注射5 ml/kg生理盐水)和染毒组(腹腔注射7.5、15和30 mg/kg ACR),每组12只,雌雄各半;步态评分评价神经行为改变;组织病理学法检测背根神经节(dorsal root ganglion,DRG)(电子显微镜)、心、肝、脾、肺、肾等(光学显微镜)的超微结构改变;神经生化法(定磷)检测大鼠背根神经节中Ca2+-ATP酶、Na+-K+-ATP酶活性的变化。结果实验结束时,30 mg/kg ACR染毒组大鼠体重较对照明显降低,步态评分较对照显著增加(P<0.05),异常体征明显;背根神经节病理改变明显,心、肝、脾、肺、肾等组织未见明显病变;丙烯酰胺各染毒组大鼠钙ATP酶(Ca2+-ATPase)、钠钾ATP酶(Na+-K+-ATPase)活性均较对照组显著降低(P<0.05)。结论背根神经节ATP酶活性的变化是丙烯酰胺神经损伤的重要生理生化基础之一。Objective This study was designed to explore neurotoxic biomarkers induced by acrylamide in rats. Methods Forty- eight wistar rats were divided into the control group and different concentrations of ACR treatment groups randomly with equal numbers of male and female. Acrylamide (7.5, 15 and 30 mg/kg) was administered to rats by intraperitoneal injection in test groups and normal saline was given to rats in the control group. The neurological and pathologic changes of dorsal root ganglion, heart, liver, spleen, lung and kidney were observed. Activities of Ca2 ~ -ATPase and Na ~ -K + -ATPase within dorsal root ganglion (DRG) were measured. Results Weight of rats in 30mg/kg ACR treatment group was decreased compared with the control group(P 〈 0. 05). Neurological changes were significant at the end of the test. Gait scores of rats in 30 mg/kg ACR treatment group were higher than that of the control group ( P 〈 0. 05 ). Pathologic changes were observable in dorsal root ganglion while no changes of heart, liver, spleen, lung and kidney were observed. Compared with the control group, the activities of Ca2+ -ATPase and NaZ* -K * -ATPase were decreased( P 〈 0. 05 ). Conclusion ATPase of dorsal root ganglion may he one of biomarkers of neurotoxicity induced by acrylamide.
