摘  要:目的探讨IL-17对人表皮角质形成细胞株(HaCaT细胞株)表达CC型趋化因子(CCL17/TARC,CCL22/MDC,CCL11/eotaxin及CCL27/CTACK)的影响。方法体外培养HaCaT细胞,加入不同浓度IL-17(20,40,80,160ng/mL)共刺激培养24h,提取总RNA并收集培养细胞上清液,分别进行实时定量RT—PCR和ELISA实验,检测CCL17,CCL22,CCL11及CCL27mRNA和蛋白水平的变化。结果20~160ng/mL的IL-17均可促进Hacarr细胞表达CCL17,CCL22及CCL11 mRNA和分泌CCL17,CCL22及CCL11,并呈剂量依赖关系(P均〈0.01);各IL-17刺激组CCL27mRNA表达水平及CCL27分泌与正常对照组间差异无统计学意义,各IL-17刺激组间两两比较差异亦无统计学意义(P均〉0.05)。结论IL-17对Hac打细胞表达CCL17,CCL22及CCL11具有明显的促进作用,为Th17细胞参与特应性皮炎发病机制提供了进一步的理论依据。Objective To explore the effect of IL-17 on CC chemokine subfamily (CCL17/TARC, CCL22/MDC, CCL11/eotaxin and CCL27/CTACK ) expression in human keratinocytes and HaCaT cells. Methods HaCaT ceils were cultured and stimulated with different concentration of IL-17 (20, 40, 80, 160ng/mL) for 24h. Real-time RT-PCR and enzyme linked immunosorbent assay were employed to detect the mRNA levels and concentrations of CCL17, CCL22, CCLll and CCL27 in HaCaT cells and cell-free supernatants, respectively. Results IL-17 (20, 40, 80, 160ng/mL) could promote the mRNA expression and secretion of CCL17, CCL22 and CCL11 in cultured HaCaT cells in a dose-dependent way( all P 〈 O. O1 ). There were no significant difference in CCL27 mRNA level and protein secretion between IL-17-stimulated group and control group, or among IL-17-stimuIated group ( all P 〉 0.05 ). Conclusion IL-17 can promote the expression of CCL17, CCL22 and CCL11 in HaCaT cells, which further substantiate the involvement of Thl7 ceils in the pathogenesis of atopic dermatitis.
