摘  要:目的研究寻常性银屑病患者红细胞补体受体1(complement receptor type 1,CR1)分子定量、密度相关基因多态性及红细胞天然免疫黏附功能(erythrocyte innate immune adhesion function,EIIAF)在寻常性银屑病发病机制中的作用。方法快速检测法检测EIIAF,改良的细胞酶联免疫分析法定量测定红细胞CR1,PCR-RFLP法检测红细胞CR1密度相关基因多态性。结果寻常性银屑病患者EIIAF显著高于正常对照组(P<0.05),患者组红细胞CR1分子定量显著高于正常对照人群(P<0.01),以上差异均有统计学意义;红细胞CR1分子定量变化与EIIAF的变化显著正相关。寻常性银屑病红细胞CR1密度相关基因多态性低表达基因(L)点突变率(17.31%)与正常对照组(10.71%)比较差异无统计学意义(P>0.05),基因多态性分布(HH:73.08%,HL:19.23%,LL:7.69%)与正常人(HH:85.71%,HL:7.14%,LL:7.14%)比较,差异无统计学意义(P>0.05)。结论寻常性银屑病患者红细胞CR1分子定量及其活性的增高不是由于红细胞CR1密度相关基因多态性变化所致。红细胞CR1分子数量及EIIAF增高可能在寻常性银屑病的发病机制中发挥重要作用。Objective To study the changes of genomic density polymorphism and quantitative expression of complement receptor type 1 ( CR1 ) on erythrocytes and erythrocyte innate immune adhesive function (EIIAF) in psoriatic patients in order to explore its possible role in the pathogeneses of psoriasis. Methods Rapid de- tection was used to detect EIIAF. The expression levels of CR1 on erythrocytes were detected by enzyme - linked immunoassay. The genomic density polymorphism of CR1 on erythrocytes was determined with poly- merase chain reaction(PCR) and Hind III restriction enzyme digestion. Results The activity of EIIAF was significantly higher in patients than in normal controls(P 〈0.05 ). The expression levels of CR1 on erythro- cytes were also higher in the patients than in normal controls(P 〈0.01 ). The levels of CR1 were positively correlated with EIIAF. Spot mutation rates of low - expressed CR1 locus on erythrocyte density gene were no significantly differences( 17.31% vs 10.71% , P 〉0.05) between the patients and the controls. The distri- bution of polymorphism of erythrocyte CR1 density gene was not significantly different ( P 〉 0.05 ) between the patients ( HH : 73.08% , HL : 19.23% , LL : 7.69% ) and healthy individuals ( HH : 85.71% , HL : 7. 14% , LL:7.14% ). Conclusion Increased the expression levels of CR1 on erythrocytes and EIIAF in psoriasis were not due to the changes in polymorphism of CR1 on erythrocyte gene. Higher CR1 and EIIAF may be involved in the pathogenesis of the psoriasis.
