摘  要:目的 观察狂犬病病毒PM株在人二倍体MRC-5细胞中的感染特性。方法 分别将0.05、0.06、0.07、0.08、0.09、0.10MOI的PM株狂犬病病毒接种至MRC-5细胞中,显微镜观察感染细胞的状态;间接免疫荧光法检测病毒收获液的滴度;小鼠NIH法测定病毒灭活液的效价。结果 MOI为0.06~0.07时,病毒收获前细胞出现聚集现象,液体中基本无死细胞;病毒收获液的滴度和病毒灭活液的效价较高。结论 观察了狂犬病病毒PM株在人二倍体MRC-5细胞中的感染特性,为疫苗生产工艺的优化提供了实验依据。Objective To observe the infectious characteristics of rabies virus PM strain in human diploid MRC-5 cells. Methods MRC-5 cells were inoculated with PM strain at MOIs of 0. 05, 0. 06, 0. 07, 0. 08, 0. 09 and 0. 10 respec- tively, and observed by microscopy. The harvested virus liquid was determined for titer by indirect IFA. The inactivated virus liquid was determined for potency by test in NIH mice. Results The cells inoculated with PM strain at MOIs of 0. 06 - 0. 07 showed aggregation before harvest of virus, and few dead cells were observed in liquid. The harvested virus liquid showed high titer, while the inactivated virus liquid showed high potency. Conclusion The infectious char- acteristics of rabies virus PM strain in human diploid MRC-5 cells was observed, which provided an experimental basis for optimization of production procedure of the rabies vaccine.
